人参皂甙Rb_2对培养人视网膜色素上皮细胞增生的抑制作用

摘　　要：目的　研究人参皂甙Rb2 (Ginsenoside Rb2 )对体外培养视网膜色素上皮细胞增生的直接抑制作用及可能机制。方法　通过活细胞计数法与MTT比色法分别检测不同浓度 (2 5 0、2 0 0、15 0、12 0、10 0、5 0、10 μg ml)Rb2 和Rb215 0 μg ml在不同作用时间 (6～ 12 0h)对RPE细胞增生及代谢的影响。 结果　Rb2 组细胞增殖受到抑制并出现细胞脱落 ,Rb2 2 0 0 μg ml具有最强的抑制效应 ,其明显抑制效应在 6h即出现 ,96h达高峰。Rb2 组A值明显低于对照组 ,RPE细胞生存率下降。结论　Rb2 可能通过阻滞钙通道降低细胞内钙浓度。Objective To investigate the effects of Ginsenoside Rb 2 on proliferation of cultured retinal pigment epithelium(RPE) cells and the probable mechanisms.Methods Cultured RPE cells were treated with Rb 2 by various concentrations(250,200,150,120,100,50,10μg/ml) and with Rb 2 150μg/ml at different times(6～120h),cells proliferation and metabolism were evaluated by cell count method and MTT colorimetric assay.Results There was inhibition on proliferation of RPE cells and some cells detached from the wall in Rb 2 group.The best concentration of Rb 2 was 200μg/ml.The significant inhibitory effect of Rb 2 occurred at 6h,and the strongest inhibition of Rb 2 occurred at 96h.The A value in Rb 2 groups was significantly lower than in control group and the survival rate of RPE cells lowered.Conclusion The results suggest that Rb 2 could inhibit the proliferation of RPE cells in a dose dependent and time dependent manner,the probable mechanism is that Rb 2 could block the calcium channel so intracellular concentration of calcium is lowered and the metabolism of cell is disturbed.
