摘  要:目的:探讨桂皮酸对NB4细胞诱导分化的作用及其机制。方法:采用光镜观察形态变化,嗜银蛋白染色检测各组细胞AgNORs颗粒数,流式细胞术检测CD11b和CD33,免疫组化测定细胞c-myc、c-fos蛋白表达。结果:桂皮酸作用NB细胞后,CINN可诱导NB4细胞向终末细胞分化,分化率较对照组差异明显(P<0.01);细胞AgNOR3颗粒数明显减少(P<0.01);CD11b表达升高,CD33表达下降;c-myc表达下降,c-fos蛋白表达升高(P<0.05)。结论:桂皮酸能使NB4细胞分化成熟,增殖能力下降,其机制可能是通过凋控NB4细胞的增殖、分化相关的基因c-myc、c-fos表达来抑制细胞增殖,诱导细胞分化。Objective: To investigate NB4 cells differentiation induced by cinnamic acid (CINN) and its mechanism. Methods : NB4 cells' morphlogy was observed by light microscopy, and the particles of AgNORS were tested by argyrophic technique; membrance CD33 CD11 antigens were analysed by flow cytometry, and expressions of gene products c-myc c-fos were tested by S-P method. Results: NB4 cells treated with cinnamic acid exhibitd a characteristics of cell differentiation ( P < 0.01 ) . The number of particles of AgNORs in nueleous markedly decreased ( P < 0.01) . Expressions of CD33 decreased and CD11b increased (P<0.05). Expressions of c-myc decreased and c-fos increased. Conclusion: CINN can induce differentiation and growth inhibition of NB4 cells. Its mechanism might berelated to modulation of gene expressions associated proliferation and differentiation.
