摘  要:目的:探讨雷公藤内酯醇(TP)对试验性色素膜视网膜炎(EAU)Th1反应相关因子的抑制作用。方法:用ELISA检测IL-12、IFN-γ水平;采用原位杂交染色法,观测TP对EAU的T淋巴细胞IL-12mRNA的表达的影响;采用电泳迁移率改变试验对EAU T淋巴细胞NF-AT活性检测。结果:①TP能有效的降低S-抗原诱导产生的EAU(P<0.05);②TP能降低EAU T淋巴细胞IL-12、IFN-γ分泌水平;③P能抑制EAT淋巴细胞IL-12 mRNA的表达;④TP抑制EAU T淋巴细胞NF-AT活性。结论:TP对EAU的免疫抑制作用可能是通过抑制与Th1反应相关的调节因子使Th1反应减弱,而使EAU的发病率明显降低。Objective: To studies the inhibition of triptolide(TP) on the correlactive regulative factor of TH, response in Experimental Autoimmune Uveoretinitis(EAU) . Methods: The levels of IFN-yand IL-12 were detected by ELISA The situ hybridization (ISH) was adapted to explore the expression of IL-12mRNA and the effects of TP in T Lymphocyte of EAU; The activation of NF-AT in T Lymphocyte of EAU was assayed by using electrophoretic mobility shift assay(EMSA) . Results:(1)The rate decreased in EAU induced by S - Ag that was treated by TP. (2)TP could decreased the levels in T lymphocytes of EAU to secrete IFN-yand IL-12. (3)TP could inhibit interleukin-12 gene expression. (4)TP could inhibit NF-AT Activity. Conclusion: TP could inhibit correlactive regulative factor of TH, response to cause incidence rate of EAU.
