丁酸钠诱导K562细胞肺耐药相关蛋白表达的初步研究

摘　　要：目的研究丁酸钠(NaB)对K562细胞肺耐药相关蛋白(LRP)表达的诱导作用。方法以K562细胞为体外模型,采用半定量逆转录聚合酶链反应(RT-PCR)检测NaB诱导前后K562细胞LRPmRNA的水平,采用流式细胞仪检测比较NaB诱导前后K562细胞LRP蛋白表达的变化。结果2mmol/L诱导后LRPmRMA水平、蛋白表达均明显增加。结论NaB诱导可使K562细胞LRPmRNA水平和蛋白质表达的增加。Objective To investigate the effect of sodium butyrate (NaB) o n the expression level of lung resistance-related pro-tein (LRP) in K562 cells. Me thods K562 cells, used as an in vitro model system, were treated with NaB to ind uce differentia-tion of the cells. LRP mRNA expression and protein levels in the cells were detected by reverse transcriptase-PCR (RT-PCR) and flow cytometry, respectively. Results A 1-day treatment with 2 mmol/L NaB induced the gene expre ssion of LRP to increase to the maximum in the cells, and a 3-day treatment cau sed the corresponding protein level to increase to the maximum. Con鄄clusion Bot h the mRNA level and the protein expression of LRP can be induced by NaB in K562 cells.
