孤啡肽对大鼠脑内内阿片肽含量的影响

摘　　要：目的 :研究侧脑室注射孤啡肽 (OFQ)对大鼠脑内内阿片肽含量的影响。方法 :大鼠 2 0只 ,随机分为 2组 ,生理盐水组 (NS) :大鼠右侧侧脑室注射NS ;孤啡肽组 :大鼠右侧侧脑室注射OFQ。采用推挽灌流的方法收集脑内导水管周围灰质腹侧区 (PAG)中的灌流液 ,用放射免疫法测定灌流液内内阿片样免疫活性物质的含量。结果 :孤啡肽组侧脑室注射OFQ后PAG灌流液内 β 内啡肽 ((- 10 .4± 3.2 )ng/L)和亮氨酸 脑啡肽样免疫活性物质((- 2 .9± 1.4 )ng/L)含量与对照组比较 ((- 0 .4± 2 .7)ng/L)和 ((2 .2± 0 .8)ng/L)均明显减少 (P <0 .0 1)。 结论Aim: To study the effect of orphanin FQ (OFQ) on the content of endogenous opioid peptides in rat brain. Methods:A total of 20 rats were randomly allocated into 2 groups.Normal saline group (NS):NS was injected into the rat right intracerebroventricular;orphanin FQ group (OFQ):OFQ was injected into the rat right intracerebroventricular. Using the techniques of push pull perfusion and radioimmuno assay,the content of endogenous opioid peptides in perfusate from PAG were detected. Results: After intracerebroventricular injection of OFQ, the content of β endorphin and L enkephalin was decreased in perfusate from PAG. Conclusion: Intracerebroventricular injection of OFQ could inhibit the release of endogenous opioid peptides in PAG.
