摘  要:目的 研究胰腺癌组织中smad4 mRNA ,TGF β1 和TGF βR1 的表达及其生物学意义。方法 采用原位杂交方法检测癌及癌旁组织的smad4 mRNA ,免疫组化方法检测TGF β1 和TGF βR1 。结果 5 3例胰腺癌smad4 mRNA ,TGF β1 和TGF βR1 表达阳性率明显地低于 2 5例癌旁上皮 (均P <0 .0 5 )。高分化腺癌smad4 mRNA ,TGF β1 和TGF βR1 表达阳性率明显高于低分化腺癌 (P <0 .0 5~ 0 .0 1) ,无转移癌明显高于转移癌 (P <0 .0 5~ 0 .0 1)。三者在胰腺癌中表达阳性率均存在密切相关系。结论 smad4 mRNA ,TGF β1 和TGF βR1 表达与胰腺癌的发生发展、生物学行为和预后有密切关系 ,检测三者表达水平对早期发现胰腺癌及指导临床治疗和评估预后可能有较重要价值。Objective To study the significance of expressions of smad_4mRNA,TGF-β_1, and TGF-βR_1 in pancreatic carcinoma(PC) . Methods Smad_4mRNA was detected by in situ hybridization. TGF-β_1 and TGF-βR_1 were detected by immunohistochemical method. Results The positive rates of smad_4mRNA,TGF-β_1 and TGF-βR_1 were singnificantly lower in 53 slices of pancreatic carcinoma than those in 25 slices of paracancerous tissue (all P< 0.05). The positive rates of smad_4mRNA,TGF-β_1, and TGF-βR_1 were significantly higher in cases of well differentiation cancer than those in poorly differentiation cancer(P<0.05~0.01).In the cases of non-metastatic cancer, the positive rates of smad_4mRNA,TGF-β_1 and TGF-βR_1 were significantly higher than those in the cases of metastasis(P<0.05~0.01).The close correlations were found between the expressions of smad_4mRNA,TGF-β_1 and TGF-βR_1 .Conclusions The expressions of smad_4mRNA,TGF-β_1,and TGF-βR_1 are related to the pathogenseis and development,biological behavior, and prognosis of pancreatic carcinoma.Detecting the expressions may have important clinical values in the early diagnosis,treatment,and evaluation of prognosis of pancreatic carcinoma.
