鼻咽癌患者血清中Epstein-Barr病毒早期抗原特异性抗体的IgA类抗独特型抗体的检测

摘　　要：用3.5％PEG(MW6000)预先处理被检血清以除掉免疫复合物及干扰抗原之后,用新建立的ELISA方法测定血清中Epstein-Barr病毒早期抗原特异性抗体的抗独特型(Anti-Id)抗体。67例鼻咽癌患者中,80％IgA类的Anti-Id抗体阳性(P/N值≥2.1),而56例正常人群除1例阳性外,其余均阴性。测定46例鼻咽癌患者血清EA/IgA滴度与Anti-Id的关系表明,Anti-Id抗体P/N值与IgA/EA滴度呈负相关(r=-0.6,P<0.001)。12例鼻咽癌病人血清中EA抗体的IgG类Anti-Id抗体则与正常人无显著差别。A simple enzyme-linked immunosorbent assay ( ELISA)has been developed to detect circulating IgA class anti-idiotypic antibody against antibody to EB virus early antigen ( EA ) in the sera of NPC patients. Specific murine anti-EA ( P54 ) monoclonal antibody was coated into 96 well polyvinyl microtiter plates and the HRPO-labelled sheep anti-human IgA antibody was used as conjugate. Anti-Id antibody in the su pernant after removing the immune complexes from sera with polyethylene glycol ( PEG ) 6000 could be detected. The positive rate of IgA class anti-Id antibody was 80% in 67 patients with NPC,and among the control, except one, all were negative. When the EB virus IgA/EA antibody titer is negative, 1:5, 1:10, and 1:20, the mean value is 2.9±1.158, 3.1±0.499, 2.6±0.466 and 1.8±0.599 respectively.These data suggest that IgA class anti-Id antibody against antibody to EB virus EA existed in the sera of NPC patients, and there is negative correlation(r=-0.6, P<0.001 ) between IgA class anti-Id antibody ( P/N) and IgA/EA antibody titer.
