子宫内膜癌组织中LIMD2的表达及临床意义

摘　　要：目的探讨子宫内膜癌组织中含LIM域2蛋白(LIMD2)的表达及临床意义。方法收集2014年1月至2017年12月经手术切除的子宫内膜癌组织89例、正常内膜组织57例及不典型增生组织61例,采用实时定量PCR(QPCR)法检测上述组织中LIMD2的表达情况,采用受试者工作特征(ROC)曲线评估组织LIMD2水平诊断子宫内膜癌的效能;分析LIMD2表达与子宫内膜癌临床病理特征的关系(年龄、淋巴结转移、FIGO分期、病理类型、组织学分级和肌层浸润深度);采用Kaplan-Meier Plotter在线数据库分析LIMD2表达与子宫内膜癌预后的关系。结果子宫内膜癌组织的LIMD2 mRNA水平为3.095±1.236,高于不典型增生组织的1.297±0.420和正常内膜组织的1.176±0.444(P<0.05),而不典型增生组织与正常内膜组织LIMD2表达水平的差异无统计学意义(P>0.05)。ROC曲线显示,组织LIMD2水平诊断子宫内膜癌的曲线下面积为0.901(95%CI:0.851~0.951),以1.918为截断值对应的约登指数最大(0.7865),灵敏度和特异度分别为78.65%和100.0%。LIMD2表达与年龄和病理类型及均无关(P>0.05),与淋巴结转移、FIGO分期、肌层浸润深度和组织学分级均有关(P<0.05)。Kaplan-Meier Plotter在线数据库分析结果发现,LIMD2表达与无复发生存期有关(HR=0.53,95%CI:0.29~0.98,P<0.05),而与总生存期无关(P>0.05)。结论子宫内膜癌组织中LIMD2高表达,且与淋巴结转移、FIGO分期、组织学分级、肌层浸润深度和复发有关,可能参与该肿瘤的发生、发展,可能成为子宫内膜癌诊断和病情预测的新型分子标记物。Objective To investigate the expression and clinical significance of LIM domain containing 2 protein(LIMD2)in endometrial cancer tissues.Methods Eighty-nine cases of endometrial cancer tissues,57 cases of normal endometrial tissues and 61 cases of atypical hyperplasia tissues were collected from January 2014 to December 2017.The expression of LIMD2 in the above tissues was detected by real-time quantitative PCR(QPCR).The efficacy of tissue LIMD2 level for the diagnosis of endometrial cancer was evaluated by receiver operating characteristic(ROC)curve.The relationship between LIMD2 expression and clinicopathological parameters of endometrial cancer(age,lymph node metastasis,FIGO stage,pathological type,histological grade and myometrial invasion depth)was analyzed.Results The level of LIMD2 in endometrial cancer tissues was 3.095±1.236,higher than 1.297±0.420 of atypical hyperplasia tissues and 1.176±0.444 of normal endometrial tissues(P<0.05).There was no significant difference in the level of LIMD2 between normal endometrium group and atypical hyperplasia group(P>0.05).ROC curve showed that the area under the curve of tissue LIMD2 level for diagnosis of endometrial cancer was 0.901(95%CI:0.851-0.951).When 1.918 was chosen as the cut-off point,the Yoden index was the largest(0.7865)with the sensitivity of 78.65%and specificity of 100.0%.The expression of LIMD2 was not correlated with age and pathological types(P>0.05),but with lymph node metastasis,FIGO stage,depth of muscle invasion and histological grade(P<0.05).The results of Kaplan-Meier Plotter online database analysis showed that LIMD2 expression was associated with relapse-free survival(HR=0.53,95%CI:0.29-0.98,P<0.05),while it was not associated with overall survival(P>0.05).Conclusion LIMD2 expression is enhanced and related to lymph node metastasis,FIGO staging,histological grade,depth of muscle invasion and recurrence of endometrial cancer.It may be involved in the occurrence and development of endometrial cancer and may become a new molecular marker
