摘 要:以矾根‘巴黎’(Heuchera'Pairs')为试材,研究了植物生长调节剂对矾根无菌苗叶柄诱导愈伤组织、愈伤组织分化不定芽以及诱导试管苗生根的影响。结果显示:矾根叶柄诱导愈伤组织的适宜培养基是MS+PIC0.5mg/L+6-BA0.5mg/L,诱导率达96.30%;愈伤组织分化不定芽的适宜培养基是MS+6-BA0.1mg/L+IAA0.05mg/L,分化率达81.25%,不定芽的增殖率达12.5;诱导试管苗生根的适宜培养基是1/2MS+IBA1.0mg/L,生根率达100%,且试管苗植株健壮、根系发达。本研究建立了以矾根叶柄为外植体的高效再生体系,对矾根优良种苗的离体快速繁殖具有重要意义,并为进一步开展矾根的遗传转化研究提供技术支撑。This work took Heuchera'Pairs'as experimental material and studied the effect of plant growth regulator on Heuchera aseptic seedling petiole callus induction,callus adventitious buds differentiation,and rooting of test-tube plantlet.The results indicated that the feasible medium for Heuchera petiole callus induction was MS+PIC 0.5 mg/L+6-BA 0.5 mg/L,and the induction rate reached 96.30%;the appropriate medium for callus adventitious buds differentiation was MS+6-BA 0.1 mg/L+IAA 0.05 mg/L,the differentiation rate reached 81.25%,and the proliferation rate of adventitious buds reached 12.5;The suitable rooting medium for test-tube plantlet was 1/2MS+IBA 1.0 mg/L,and the rooting rate reached 100%,additionally,the test-tube seedlings were robust and developed in root system.In this study,a highly efficient regeneration system was established with the petiole of Heuchera as explants,which was of great significance for rapid propagation in vitro of Heuchera seedlings,providing technical supports for further research on genetic transformation of Heuchera.
