摘 要:消化道内数以亿计的微生物与上皮细胞及免疫细胞等共同构成人体特有的肠道微环境。在肠管中,免疫系统的促炎和修复信号之间的平衡对于维持肠稳态必不可少。包括Toll样受体(Toll-like receptor,TLR)在内的多种模式识别受体通过识别各类病原相关分子模式及损伤相关分子模式,参与机体抗病原体的固有免疫防线。近年来研究证实,TLR信号通路在肠癌发生和发展中发挥重要作用,肠道菌群-TLR信号通路-炎性反应及肿瘤级联反应被认为是消化系统恶性肿瘤可能的发病机制之一。本文就TLR家族的结构及功能、TLR信号通路及其与肠癌发生和发展的关系进行综述。Hundreds of millions of gut microbiota,intestinal epithelial cells and immune cells together constitute a unique gastrointestinal microenvironment.In intestine,the balance between proinflammatory and repair signals of the immune system is essential for the maintenance of intestinal homeostasis.A variety of pattern recognition receptors,including Toll-like receptors(TLR),constitute the body's innate immune defense against pathogens by identifying pathogenassociated molecular patterns and damage-associated molecular patterns.Many investigations support that TLR signaling pathway plays an important role in the genesis and development of intestinal malignancies.Intestinal flora-TLR signaling pathway-inflammation and cancer cascade reaction are considered as the possible pathogenesis of colorectal cancer.The structure and function of TLR family,TLR signaling pathway and its role in the genesis and development of colorectal cancer will be summarized in this review.
