摘　　要：目的研究卵巢癌细胞系中的肿瘤干细胞(CSC)样细胞一侧群细胞(SP)中不同亚群细胞的干性是否有差异,分析CSC的异质性。方法选择人浆液性卵巢癌顺铂耐药细胞株C13,采用双苯酰亚胺H0chest33342染色,应用流式细胞仪荧光激活分选法获得高度排斥染色的侧群细胞(LSP)、低度排斥染色的侧群细胞(USP)及能被双苯酰亚胺HocheSt33342染色的非侧群细胞(NSP),体外试验检测3个不同亚群的细胞体外成球能力、表达干细胞标志物、对高浓度顺铂的耐药性是否有差异。结果人卵巢癌细胞C13存在具有干性特征的SP,其亚群LSP比USP表达更高水平干细胞标志物,具有更高的体外成球能力,对顺铂具有更高的耐药性,差异有统计学意义。NSP不具有干性特征。结论体外试验证明卵巢癌细胞的SP不同亚群细胞具有不同的干性特征,CSC具有异质性。Objective To explore the stemness of different subtypes in human ovarian cancer stem cell( CSC)-like side population cells( SP),analyze the heterogeneity of CSC. Methods Human ovarian cancer C13 cells were collected,bisbenzimide Hochest33342 staining was performed,flow cytometry and fluorescence-activated cell sorting were used to obtain LSP cells( high rejection of staining),USP cells( moderate rejection of staining),and NSP cells( stained by bisbenzimide Hochest33342). In vitro test was used to detect the capability of sphere-forming,the expression of stem cell markers,and drug resistance to cisplation at a high drug concentration among the three groups. Results SP cells with stemness were observed among human ovarian cancer C13 cells,compared with USP,LSP cells had higher expression levels of stem cell markers,higher capability of sphere-forming,and higher drug resistance to cisplation,there were statistically significant differences. NSP cells had no stemness. Conclusion In vitro test showed that different subtypes in human ovarian cancer SP cells have different stemness.
