摘 要:目的观察纤维蛋白胶(FG)复合脱蛋白小牛骨(DBBM)在兔颅骨极限骨缺损模型中的成骨效果。方法构建两种DBBM/FG复合材料,分别为Bio-oss/FG与HA/FG。将24只新西兰兔按随机数字表法分为Bio-oss组、Bio-oss/FG组、HA组、HA/FG组、FG组和空白组,每组4只。构建兔颅骨极限骨缺损模型行骨修复手术,按照分组植入相应材料。术后4、8周分批处死新西兰兔,取出骨缺损区及其组织,各组骨片标本行微计算机断层扫描(Micro-CT)及硬组织切片Mc-Neal染色观察成骨效果。结果术后4周时,Bio-oss/FG组的成骨效果优于Bio-oss组,HA/FG组的成骨效果优于HA组,且以上4组的成骨效果均优于FG组和空白组,差异均有统计学意义(均P<0.05)。而术后8周时,Bio-oss/FG组与Bio-oss组、HA/FG组与HA组的成骨效果比较差异均无统计学意义(均P>0.05)。FG在4周时明显促进了DBBM组的成骨作用,但8周时与单纯应用DBBM组无统计学差异,单纯应用FG不能促进成骨。结论FG复合DBBM修复骨缺损,可较好的填补DBBM塑形困难的缺陷,在术后早期有促进成骨作用。Objective To evaluate the osteogenic effect of fibrin glue(FG)combined with two kinds of deproteinized bovine bone mineral(DBBM)in the skull of rabbits.Methods Two kinds of DBBM/FG(Bio-oss/FG and HA/FG)composite materials were fabricated.Twenty four New Zealand white rabbits were randomly divided into 6 groups with 4 in each group.The critical bone defect model was fabricated in the skull of rabbits,and the defected skulls were repaired with Bio-oss,Bio-oss/FG,HA,HA/FG or FG,respectively.The rabbits with unrepaired skull were used as control group.Bone samples were taken 4 and 8 weeks after surgery and the skull healing was detected by Micro-CT and Mc-Neal staining.Results Four weeks after surgery,the osteogenic effect in Bio-oss/FG group was significantly higher than that in Bio-oss group;the osteogenic effect in HA/FG group was higher than that in HA group;and the effect of above four groups were all higher than that in FG group and the control group(P<0.05).Eight weeks after surgery,there was no significant difference in osteogenic effect between Bio-oss/FG group and Bio-oss group,HA/FGgroup and HA group(P>0.05).Conclusion FG combined with DBBMcould overcome the difficulties in shaping and promote osteogenesis in the early period of bone healing.
