摘 要:目的分析中期因子(MK)在绒毛膜癌(以下简称绒癌)中的表达情况,探讨MK表达与绒癌病理特征的关系。方法培养人绒癌细胞株JEG-3、JAR,选择对数生长期的JEG-3、JAR细胞进行实验,采用Matrigel体外侵袭试验检测细胞体外侵袭能力,RT-PCR法检测细胞MK mRNA表达水平,Western blot法检测MK蛋白表达水平。选取32例绒癌患者的绒癌组织标本,采用免疫组织化学法检测绒癌组织MK蛋白表达情况,分析其与临床病理特征的关系,包括患者年龄、肿瘤临床分期、国际妇产科联盟(FIGO)预后评分等。结果JEG-3细胞穿膜数明显多于JAR细胞(P<0.05),即JEG-3细胞体外侵袭能力强于JAR细胞。培养12、24、48h时,JEG-3细胞MK mRNA表达水平均高于低侵袭性JAR细胞(均P<0.05)。JEG-3细胞MK蛋白表达水平高于JAR细胞(P<0.05)。绒癌组织中MK蛋白表达阳性率在<40岁与≥40岁患者间的差异无统计学意义(P>0.05),临床分期为Ⅲ~Ⅳ期的患者高于Ⅰ~Ⅱ期的患者(P<0.05),FIGO预后评分高危的患者高于低危的患者(P<0.05)。结论MK在绒癌组织中高表达,且与绒癌细胞侵袭能力有关,其表达阳性率随绒癌分期的升高而增高,可作为判断绒癌恶性程度和预后的重要生物标志物。Objective To analyze the expression of midkine(MK)in choriocarcinoma and its relationship with clinicopathological features of patients.Methods The expression of protein and MK mRNA in human choriocarcinoma JEG-3 and JAR cells were detected with Western blot and RT-PCR,respectively.The cell invasion ability in vitro was examined with Matrigel invasion test.The tissue samples of 32 patients with choriocarcinoma were collected,and the expression of MK protein in choriocarcinoma tissue was detected by immunohistochemistry,its relationship with clinicopathological characteristics of patients was analyzed.Results The number of membrane penetration of JEG-3 cells was significantly higher than that of JAR cells(P<0.05).When cultured for 12,24 and 48h,the expression levels of MK protein and mRNA in highly invasive JEG-3 cells were all higher than those in low-invasive JAR cells(all P<0.05).The expression of MK protein in the choriocarcinoma tissue was not corrected with patients age(P>0.05).The positive rate of MK protein expression in clinical stageⅢ~Ⅳcancer was significant higher than that of stageⅠ~Ⅱcancer(P<0.05);the positive rate of MK protein expression in patients with high FIGO prognostic score was higher than that in those with lowFIGOscore(P<0.05).Conclusion MK is highly expressed in choriocarcinoma tissues and it is related to the invasion ability of choriocarcinoma cells.Its expression increases with the increase of choriocarcinoma stage,indicating that it may be used as a biomarker for malignancy degree and prognosis of choriocarcinoma.
