摘  要:目的研究头颈部鳞状细胞癌Tca8113和HSC2细胞系暴露于2%异氟醚后其生物学行为发生改变的特点,以期为头颈鳞状细胞癌手术麻醉方式提供细胞水平的理论指导。方法头颈部鳞状细胞癌细胞系用2%异氟醚分别作用于Tca8113和HSC2细胞系3 h、6 h后,体外培养72 h,用MTT法检测2%的异氟醚对Tca8113和HSC2细胞的增殖能力的影响。用Transwell小室培养24 h检测2%异氟醚对细胞的侵袭能力的影响。结果MTT法检测和Transwell小室实验提示,接受2%异氟醚暴露3~6 h后,Tca8113和HSC2细胞系与没有接受异氟醚暴露的对照组相比,其增殖能力与侵袭能力均显著增强,差异具有统计学意义(P<0.05)。结论异氟醚增加了头颈部鳞状细胞肿瘤Tca8113和HSC2细胞系在体外的恶性程度,提示使用异氟醚静吸复合麻醉可能增加头颈部肿瘤细胞增殖和转移的能力。Objective To study the characteristics of biological behavior changes of Tca8113 and HSC2 cell lines in head and neck squamous cell carcinoma after exposed to isoflurane in vitro,so as to provide theoretical guidance for the surgery.Methods Tca8113 and HSC2 cell lines were cultured in vitro for 72 hours after exposed to 2.0%isoflurane 3 hours and 6 hours respectively.MTT test was used to detect the effect of 2%isoflurane on cell proliferation.Transwell test was used to detect the effect of 2%isoflurane on cell invasion ability.Results MTT and Transwell indicated that the proliferation and invasion ability of Tca8113 and HSC2 cell lines showed significant increase after exposed to 2.0%isoflurane 3 to 6 hours,and the differences were statistically significant compared with the control group without isoflurane exposure(P<0.05).Conclusion Isoflurane can increase the malignancy of head and neck squamous cell carcinoma cell lines in vitro.It is suggested that isoflurane might enhance tumor development and promote metastasis of tumor cells in patients with head and neck squamous cell carcinoma.
