摘 要:目的探讨高压氧联合化疗治疗食管癌模型裸鼠的疗效,及对磷酸酶及张力蛋白同源基因(PTEN)、缺氧诱导因子-1α(HIF-1α)表达的影响。方法选取30只食管癌模型裸鼠,采用随机数字表法将裸鼠分为模型组、化疗组和联合组,每组10例;另选取10只同期饲养的健康裸鼠为对照组。对照组与模型组以生理盐水灌服;化疗组灌服替加氟溶液;联合组在化疗组的基础上给予高压氧(HBO)治疗。对3组食管癌模型裸鼠治疗后的肿瘤体积进行比较,并对4组裸鼠中PTEN、HIF-1α的表达水平进行比较。结果化疗组与联合组裸鼠的肿瘤体积均小于模型组,联合组裸鼠的肿瘤体积小于化疗组,差异均有统计学意义(P﹤0.05)。模型组、化疗组和联合组裸鼠的移植瘤组织中PTEN的表达水平均低于对照组裸鼠的正常食管黏膜组织,HIF-1α的表达水平均高于对照组裸鼠的正常食管黏膜组织(P﹤0.05);模型组裸鼠的移植瘤组织中PTEN的表达水平低于化疗组和联合组,HIF-1α的表达水平高于化疗组和联合组(P﹤0.05);化疗组裸鼠的移植瘤组织中PTEN的表达水平低于联合组,HIF-1α的表达水平高于联合组(P﹤0.05)。结论HBO联合化疗可以上调食管癌裸鼠模型瘤体中PTEN的表达,同时下调HIF-1α的表达,提高化疗药物对食管癌的治疗效果。Objective To investigate the curative effect of hyperbaric oxygen combined with chemotherapy on esophageal cancer in nude mice and the influence on the expression of gene of phosphate and tension homolog(PTEN)and hypoxia-inducible factor-1α(HIF-1α).Method Thirty nude mice with esophageal cancer were randomly divided into model group,chemotherapy group and combined therapy group,with 10 in each;and then another 10 healthy nude mice fed during the same period were used as control.The control group and model group were given normal saline,the chemotherapy group was additionally given tegafur solution,and the combined therapy group was administered with hyperbaric oxygen therapy in addition to the regimen prescribed for chemotherapy group.The tumor volume and expression levels of PTEN and HIF-1αwere compared after treatment in nude mice of each group.Result After treatment,the volume of tumors in chemotherapy group and combined therapy group significantly lesser than that in model group(P<0.05),and the volume of tumors in combined therapy group was significantly less than that in chemotherapy group(P<0.05).Transplanted tumor tissues from nude mice in model group,chemotherapy group and combined therapy group had significantly lower expression level of PTEN but higher expression level of HIF-1αcompared to normal esophageal mucosa of nude mice in control group(P<0.05);while the PTEN expression level was significantly lower in the transplanted tumor tissues of model group than that in chemotherapy group and combined therapy group,and lower HIF-1αexpression level was higher instead(P<0.05);the combined therapy group had higher expression level of PTEN,and lower HIF-1αexpression level than that in chemotherapy group(P<0.05).Conclusion Hyperbaric oxygen combined with chemotherapy can increase the expression of PTEN and decrease the expression of HIF-1αin nude mice with esophageal cancer,and improve the effect of chemotherapeutic drugs on esophageal cancer.
