摘　　要：目的探讨WNT1诱导信号通路蛋白2(WISP2)在非小细胞肺癌(NSCLC)中的表达情况及对肺癌A549细胞周期分布和凋亡情况的影响。方法取60例NSCLC患者的NSCLC组织和相应的癌旁正常组织,采用免疫组化法检测WISP2蛋白的阳性表达情况及与NSCLC患者临床特征的关系。将WISP2蛋白过表达的载体转染至肺癌A549细胞,作为观察组,未转染的肺癌A549细胞作为对照组,采用流式细胞仪检测两组细胞周期分布情况及凋亡情况。结果NSCLC组织中WISP2蛋白的阳性表达率为51.67%,低于癌旁正常组织的70.00%,差异有统计学意义(P﹤0.05)。不同吸烟情况、病理类型、临床分期、分化程度、淋巴结转移情况NSCLC患者NSCLC组织中WISP2蛋白阳性表达率比较,差异均有统计学意义(P﹤0.05)。观察组肺癌A549细胞G1期细胞比例明显高于对照组细胞,S期和G2期细胞比例均明显低于对照组细胞,差异均有统计意义(P﹤0.01)。观察组细胞凋亡率为(5.55±0.69)%,明显高于对照组细胞的(3.05±0.43)%,差异有统计意义(P﹤0.01)。结论WISP2在NSCLC组织中表达水平较低,其在NSCLC的发生发展过程中发挥重要作用。Objective To study the expression of WNT1 inducible signaling pathway protein 2(WISP2)in nonsmall cell lung cancer(NSCLC)and its effect on the cell cycle distribution and cell apoptosis of lung cancer cells A549.Method Sixty patients with NSCLC were included,from whom the NSCLC tissues and adjacent normal tissues were collected,the expression of WISP2 was investigated using immunohistochemical assay,and the relationship with patients’clinical characteristics were explored.The vector overexpressing WISP2 protein was transfected into lung cancer A549 cells,as the observation group,untransfected lung cancer A549 cells were used as the control group.Flow cytometry was utilized to detect the distribution of cell cycle and cell apoptosis in the two groups.Result The positive expression rate of WISP2 in NSCLC tissues was 51.67%,which was significantly lower than that in the control group at 70.00%.The positive expression rates of WISP2 protein in NSCLC patients with different smoking status,pathological type,clinical stage,differentiation degree and lymph node metastasis were statistically significant(P<0.05).In observation group,the proportion of A549 cells in G1 phase was significantly higher compared to control group,while those for phase S and phase G2 were lower instead(P<0.01).The apoptosis rate in observation group was(5.55±0.69)%,which was significantly higher compared to the(3.05±0.43)%in control group,the difference was of statistical significance(P<0.01).Conclusion WISP2 is with relatively low expression in NSCLC,indicating the essential role in the occurrence and development of NSCLC.
