摘 要:采用负重爬梯和跑台模式分别进行抗阻运动和有氧运动干预,研究不同运动方式对不同月龄雄性SAMP8小鼠腓肠肌组织氧化应激水平的差异变化。3月龄和6月龄的健康雄性小鼠随机分组,进行8周训练后检测各组小鼠腓肠肌总抗氧化能力(T-AOC)、总超氧化物歧化酶(T-SOD)和过氧化氢酶(CAT)活性及丙二醛(MDA)活性。结果显示,老年对照组T-AOC、T-SOD、CAT活性显著低于青年对照组(P<0.05)。两种干预方式均可导致青年小鼠T-AOC、T-SOD显著降低,其中T-AOC降幅83%和95%,T-SOD降幅70%和66%。而老年小鼠在运动干预后显著升高,其中T-AOC增幅429%和105%,T-SOD增幅458%和342%。CAT指标表现出运动方式上的差异,青年抗阻运动组显著高于对照组(P<0.05),而有氧运动组无显著差异。MDA指标显示,抗阻与有氧干预均可促使青年和老年小鼠腓肠肌中MDA的生成,但老年抗阻运动组MDA含量显著高于有氧运动组(P<0.05)。研究表明,小鼠腓肠肌中氧化应激水平与运动方式和年龄密切相关,运动干预后,青年和老年小鼠缓解氧化损伤的策略不同,青年小鼠通过体内较强抗氧化能力清除自由基,而老年小鼠则通过防御系统的被动诱导上调缓解氧化应激,提示中老年人群适量的有氧运动有利于增强抗氧化能力以缓解体内自由基引起的氧化损伤。The weight-bearing climb ladder and animal treadmill models was adopted to carry out resistance exercise and aerobic-exercise to explore the divergence change of different sports on gastrocnemius muscle oxidative stress levels of SAMP8 mice at different ages,three-month-old and six-month-old healthy male mice were randomly divided into different group.After 8 weeks of training,the activity of the total antioxidant capacity(T-AOC),superoxide dismutase(T-SOD),catalase(CAT),and malondialdehyde(MDA)content in gastrocnemius muscle of mouse hindlimb were detected.The results showed that T-AOC,(TSOD)and CAT activities in the old control group were significantly lower than those in the young control group(P<0.05).T-AOC and(T-SOD)were significantly reduced(P<0.05)in young mice by both intervention methods,in which T-AOC decreased by 83% and 95%,and(T-SOD)decreased by 70% and66%.However,those in old mice were significantly increased(P<0.05)after exercise intervention,in which T-AOC increased by 429% and 105%,and(T-SOD)increased by 458% and 342%.CAT index showed the difference in exercise style,and the youth resistance exercise group was significantly higher than the control group(P<0.05),while there was no significant difference in the aerobic exercise group.MDA indexes showed that both resistance and aerobic intervention could promote the generation of MDA in the gastrocnastus muscle of young and old mice,but MDA content in the elderly resistance exercise group was significantly higher than that in the aerobic exercise group(P<0.05).Moreover,the results showed that the level of oxidative stress in the gastrocnemius muscle of mice was closely related to exercise pattern and age.The young mice scavenged free radicals through strong antioxidant capacity in vivo,while old mice alleviated oxidative stress through passive up-regulation induced by defense system.It is suggested that moderate aerobic exercise in middle-aged and senior people is beneficial to enhance antioxidant capacity to alleviate oxidative damage caused by fre
