保肺定喘汤含药血清对烟草烟雾提取物诱导大鼠肺动脉平滑肌细胞增殖作用的影响

摘　　要：目的:探讨保肺定喘汤含药血清对烟草烟雾提取物(CSE)诱导肺动脉平滑肌细胞(PASMCs)增殖抑制的干预作用。方法:原代培养大鼠远端PASMCs,分为5组:正常对照组、模型组(10%CSE)、空白血清组、低剂量含药血清组(10%CSE+10%含药血清)和高剂量含药血清组(10%CSE+20%含药血清)。MTT法检测各组细胞增殖情况;流式细胞术(FCM)检测各组细胞周期情况;WesternBlot法检测各组细胞增殖细胞核抗原(PCNA)表达。结果:CSE能刺激PASMCs细胞增殖(P<0.01,P<0.05),使细胞周期G1期百分比下降(P<0.01),S期的百分比增加(P<0.01),PCNA含量升高(P<0.01);保肺定喘汤含药血清能抑制CSE诱导的细胞增殖,升高细胞周期G1期百分比(P<0.01),降低S期的百分比及PCNA含量(P<0.01)。结论:保肺定喘汤含药血清能有效减轻CSE诱导PASMCs增殖,其机制可能通过抑制PASMCs的PCNA表达。Objective:To observe the effects of Baofei Dingchuan Decoction medicated serum on proliferation of rat pulmonary arterial smooth muscle cells(PASMCs)by cigarette smoke extract.Methods:Rat distal PASMCs were isolated and primarily cultured.The cells were divided into 5 groups:blank control group,model group blank serum group,low dose medicated serum group(10%CSE+10%medicated serum)and high dose medicated serum group(10%CSE+20%medicated serum).Cell proliferation were determined by MTT.The cell cycle were detected by flow cytometry and the protein expression of PCNA were detectrd by Western Blot.Results:CSE promoted the proliferation of PASMCs(P<0.01,P<0.05),decreased the percentage of G1 cells(P<0.01),incrased the percentage of S cells and the protein expression of PCNA(P<0.01).Baofei Dingchuan Decoction medicated serum could reduce the proliferation of PASMCs,increase the percentage of G1 cells(P<0.01),decrase the percentage of S cells and the protein expression of PCNA(P<0.01).Conclusion:Baofei Dingchuan Decoction medicated serum might reduce the CSE-indused proliferation of PASMCs by adjusting the expression of PCNA.
