摘  要:目的阐明miR-26b对子宫内膜癌细胞迁移和侵袭的影响及机制。方法在子宫内膜癌Ishikawa细胞中转染miR-26b mimics,real time PCR方法检测miR-26b表达,Transwell小室测定细胞迁移和侵袭能力变化,Western blot检测细胞中E-cadherin、vimentin、β-catenin、c-myc蛋白表达。用Wnt信号激活剂处理转染miR-26b mimics后的子宫内膜癌细胞,采用上述方法检测细胞迁移和侵袭。Western blot检测细胞中E-cadherin、vimentin及Wnt信号通路相关蛋白β-catenin、c-myc的表达。结果转染miR-26b mimics后的子宫内膜癌细胞中miR-26b水平升高(P<0.05),细胞侵袭和迁移能力下降(P<0.05),E-cadherin蛋白表达水平升高(P<0.05),vimentin蛋白表达水平降低(P<0.05),β-catenin、c-myc蛋白表达减少(P<0.05)。Wnt信号激活剂可以逆转上调miR-26b对子宫内膜癌细胞侵袭、迁移和E-cadherin、vimentin、β-catenin、c-myc蛋白表达影响。结论miR-26b降低子宫内膜癌细胞侵袭和迁移能力,作用机制与抑制Wnt信号有关。Objective To elucidate the effect and mechanism of miR-26b on the migration and invasion of endometrial cancer cells.Methods After miR-26b mimics was transfected into endometrial cancer Ishikawa cells,real time PCR was used to detect the miR-26b level,Tanswell was used to detect the cell migration and invasion of Ishikawa cells,and Western blot was used to detect the expression of E-cadherin,vimentin,β-catenin and c-myc.Endometrial cancer cells transfected with miR-26b mimics were treated with Wnt signaling activators,and then the cell migration and invasion were detected using Tanswell,and the expression of E-cadherin,vimentin and Wnt signaling pathway-related proteinsβ-catenin and c-myc was detected by Western blot.Results The level of miR-26b in endometrial cancer cells transfected with miR-26b mimics was increased(P<0.05),the abilities of cell invasion and migration were decreased(P<0.05),the expression of E-cadherin protein was increased(P<0.05),and the protein expression of vimentin,β-catenin and c-myc protein decreased(P<0.05).Wnt signal activator reversed the effects of up-regulation of miR-26b on invasion,migration and expression of E-cadherin,vimentin,beta-catenin and c-myc in endometrial cancer cells.Conclusion The miR-26b may reduce the invasion and migration of endometrial cancer cells,which is related to the inhibition of Wnt signal.
