摘 要:目的检测首发精神分裂症(FES)患者血清中miR-103a、脑源性神经营养因子(BDNF)的表达情况,探讨二者在FES中的作用及关系。方法选取2016-08~2017-09本院69例FES患者作为研究对象,同期选取70例健康体检者作为对照组。应用实时荧光定量聚合酶链式反应(qRT-PCR)法检测血清中miR-103a表达水平,应用酶联免疫吸附(ELISA)法检测血清中BDNF表达水平,获得阳性与阴性症状量表(PANSS)、威斯康星卡片分类测验(WCST)评分,并分析FESF患者血清中miR-103a与BDNF相关性及两者与PANSS评分、WCST评分的相关性。结果两组一般资料比较,差异无统计学意义(P>0.05),FES组患者血清中miR-103a水平比对照组高,BDNF水平比对照组低(P<0.05),两者呈负相关(P<0.05)。FES患者miR-103a水平与PANSS评分中攻击危险性、WCST评分中错误应答数、非持续性错误数呈正相关(P<0.05),BDNF水平与PANSS评分中PANSS阴性、WCST评分中错误应答数、持续性错误数、非持续性错误数呈负相关(P<0.05)。结论在两组一般资料无明显差异情况下,FES患者血清中miR-103a高表达、BDNF低表达,两者呈负相关,且两者与认知能力相关指标关系密切。Objective To detect the expression of miR-103a and brain-derived neurotrophic factor(BDNF)in serum of patients with first-episode schizophrenia(FES),and to explore their role and relationship in FES.Methods Totally 69 patients with FES in our hospital from August 2016 to September 2017 were selected as FES group,and 70 healthy subjects were selected as control group.Real-time quantitative reverse transcription-polymerase chain reaction(qRT-PCR)was used to detect the expression of miR-103a in serum.Enzyme-linked immunosorbent assay(ELISA)was used to detect the expression of BDNF in serum.Positive and negative symptoms scale(PANSS)and Wsiconsin card sorting test(WCST)scores were obtained.The correlation between serum miR-103a and BDNF in patients with FESF,and the correlation between them and PANSS score,WCST score were analyzed.Results There was no significant difference in the general data between the two groups(P>0.05).The level of serum miR-103a in FES group was higher than that in control group,and the level of BDNF was lower than that in control group(all P<0.05).The level of miR-103a was negatively correlated with the BDNF,and the level of miR-103a was positively correlated with the risk of aggression in PANSS score,response errors and non-persistence errors in WCST score of FES patients(all P<0.05),while the level of BDNF was negatively correlated with PANSS negative in PANSS score,response errors,perseverative errors and non-persistence errors in WCST score of FES patients(P<0.05).Conclusion Based on no significant difference in general information between the two groups,the expression of miR-103a is high and the expression of BDNF is low in serum of patients with FES,and they are negatively correlated and closely related to cognitive competence-related indicators.
