摘  要:目的研究补肾壮骨颗粒对去势大鼠骨密度和血清、骨髓组织中类固醇受体辅助激活因子3(SRC-3)及过氧化物酶体增殖物活化受体γ协同刺激因子-1α(PGC-1α)表达的影响。方法将40只大鼠随机分为假手术组、模型组、中药组、阿仑膦酸钠组,每组10只。模型组、中药组、阿仑膦酸钠组均建立去卵巢骨质疏松模型,造模4 d后,中药组给予补肾壮骨颗粒2.5 mg/kg灌胃,阿仑膦酸钠组给予阿仑膦酸钠7 mg生药/kg灌胃,假手术组和模型组给予10 mL/kg生理盐水灌胃。连续干预12周后取血,采用酶联免疫吸附法(ELISA)检测4组大鼠血清SRC-3及PGC-1α水平;处死大鼠,运用小动物双能X射线骨密度仪测量4组大鼠股骨离体骨密度(BMD),实时荧光定量PCR法检测4组大鼠骨髓组织中SRC-3 mRNA及PGC-1αmRNA表达量。结果模型组、中药组、阿仑膦酸钠组血清SRC-3、PGC-1α水平及SRC-3 mRNA、PGC-1αmRNA表达量均明显低于假手术组(P均<0.05),中药组均明显高于模型组和阿仑膦酸钠组(P均<0.05);阿仑膦酸钠组SRC-3 mRNA表达量明显高于模型组(P<0.05),而血清SRC-3、PGC-1α水平和PGC-1αmRNA表达量与模型组比较差异均无统计学意义(P均>0.05)。中药组和模型组大鼠骨密度均明显低于假手术组(P均<0.05),中药组和阿仑膦酸钠组骨密度均明显高于模型组(P均<0.05),中药组与阿仑膦酸钠组比较差异无统计学意义(P>0.05)。结论补肾壮骨颗粒可能通过提高去势大鼠血清及骨髓组织中SRC-3和PGC-1α分子及基因表达水平而促进骨形成,从而提高骨密度。Objective It is to study the effect of Bushen Zhuanggu Granule on bone mineral density,and the expression of steroid receptor coactivator 3(SRC-3)and peroxisome proliferator-activated receptorγco-stimulatory factor-1α(PGC-1α)in serum and bone marrow tissue of ovariectomized rats.Methods Forty rats were randomly divided into sham operation group,model group,Chinese medicine group and alendronate group,with 10 rats in each group.The ovariectomized osteoporosis models were all established in model group,the Chinese medicine group and the alendronate group.In 4 days after modeling,the Chinese medicine group was given Bujiu Zhuanggu Granules 2.5 mg/kg by intragastrical administration,and the alendronate group was given alendronate sodium 7 mg crude drug/kg by intragastrical administration,and sham operation group and model group were given 10 mL/kg normal saline by intragastrical administration.After 12 weeks of continuous intervention,the blood of the rats was taken to detect the serum levels of SRC-3 and PGC-1αby enzyme-linked immunosorbent assay(ELISA).Then the rats were sacrificed and their bone mineral density(BMD)of femur was detected bt the small animal dual-energy X-ray absorptiometry.The expression of SRC-3 mRNA and PGC-1αmRNA in bone marrow tissue of rats were detected by real-time fluorescent quantitative PCR.Results The levels of serum SRC-3,PGC-1αand SRC-3 mRNA and PGC-1αmRNA in the model group,Chinese medicine group and alendronate group were significantly lower than those in the sham operation group(P<0.05),and these indexes in the Chinese medicine group were significantly higher than those in the model group and the alendronate group(P<0.05),the expression of SRC-3 mRNA in the alendronate group was significantly higher than that in the model group(P<0.05),while there was no significant difference in the expression of serum SRC-3,PGC-1αlevel and PGC-1αmRNA between the model group and the alendronate group(P>0.05).The bone mineral density of the Chinese medicine group and the model group were si
