摘　　要：目的:探讨pcDNA3.1-Crisp-1DNA疫苗的生育抑制效果。方法:选取6~8周龄SPF级BALB/c小鼠40只雌雄各半,随机将雌性小鼠分为观察A组和对照A组,雄性小鼠分为观察B组和对照B组每组10只,观察组经肌肉接种重组质粒pcDNA3.1-Crisp-1,对照组接种pcDNA3.1空载体,酶联免疫吸附试验检测血清富含半胱氨酸分泌蛋白-1(Crisp-1)水平;同时选取40只正常小鼠行避孕效果试验,HE染色观察卵巢、睾丸、附睾组织。结果:观察A组和观察B组血清Crisp-1抗体OD值(0.673±0.102、0.680±0.101)均高于对照A组和对照B组,小鼠妊娠率(30.0%、20.0%)低于对照A组和对照B组,每窝产仔数(5.6±0.3只、5.5±0.9只)低于对照A组和对照B组(均P<0.05);观察A组和对照A组小鼠卵巢组织结构正常,无萎缩。观察B组和对照B组小鼠睾丸、附睾基底膜完整,曲细精管直径正常,睾丸内有丰富的精子和各级精细胞,附睾有大量成熟精子。结论:pcDNA3.1-Crisp-1DNA疫苗有一定抑制生育作用,值得进一步研究。Objective:To investigate the reproductive inhibition effect of pcDNA3.1Crisp1 DNA vaccine.Methods:20 female and 20 male SPF BALB/c mice aged 6-8 weeks were randomly divided into observation group A,observation group B,control group A,and control group B(10 mice in each group).The mice in the observation group A and B were muscle injected with recombinant plasmid pcDNA3.1 Crisp1,and the mice in the control group A and B were muscle injected with empty vector of pcDNA3.1 The serum Crisp 1 level of all mice were detected by ELISA.During the same time,the study of contraceptive effect was conducted in these normal mice,and their ovarian,testis and epididymis tissues were observed after HE staining.Results:The OD antibody levels of serum Crisp 1 of mice in the observation group A and B were(0.673±0.102)and(0.680±0.101),which were significant higher than those of mice in the control group A and B(P<0.05).The pregnancy rates of mice in the observation group A and B were 30.0%and 20.0%,which were significant lower than those of mice in the control group A and B(P<0.05).The number of baby mice in the observation group A and B was(5.6±0.3)pieces and(5.5±0.9)pieces,which were significant less than those of mice in the control group A and B(P<0.05).The ovarian tissues structures of all female mice were normal without atrophy.As for all male mice,the basement membrane of testis and epididymis were intact,and the diameter of seminiferous duct was normal,there were abundant sperm in testis and sperm cells,and there were plenty of mature sperm in epididymis.Conclusion:pcDNA3.1Crisp1 DNA vaccine has some inhibiting effect on fertility of mice,which should be researched further.
