摘 要:目的探讨2-甲氧雌二醇(2-ME2)对鼻咽癌干细胞增殖、迁移及放疗抵抗性的影响。方法选取鼻咽癌CNE-2细胞构建鼻咽干细胞模型,采用随机数字表法分为0μmol/L组、4μmol/L组和8μmol/L组。采用CCK-8细胞增殖实验检测各组细胞增殖情况,采用Transwell小室迁移实验检测各组细胞迁移情况,采用克隆形成实验检测细胞放疗抵抗性,采用Western blot检测核因子-κB(NF-κB)p65和低氧诱导因子(HIF-1α)蛋白表达。结果4μmol/L组和8μmol/L组细胞OD值明显低于0μmol/L组(P<0.05);8μmol/L组细胞OD值明显低于4μmol/L组(P<0.05)。4μmol/L组和8μmol/L组穿膜细胞数明显少于0μmol/L组(P<0.05);8μmol/L组穿膜细胞数明显少于4μmol/L组(P<0.05)。4μmol/L组和8μmol/L组细胞0、8 Gy照射后克隆形成能力明显低于0μmol/L组(P<0.05);8μmol/L组细胞0、8 Gy照射后克隆形成能力明显低于4μmol/L组(P<0.05)。4μmol/L组和8μmol/L组细胞NF-κB p65和HIF-1α蛋白表达明显低于0μmol/L组(P<0.05);8μmol/L组细胞NF-κB p65和HIF-1α蛋白表达明显低于4μmol/L组(P<0.05)。结论2-ME2能抑制鼻咽癌干细胞增殖及迁移能力,同时降低其放疗抵抗性,机制可能与其抑制HIF-1/NF-κB信号通路有关,值得进一步研究。Objective To investigate the effect of 2-methoxyestradiol(2-ME2)on proliferation,migration and resistance to radiotherapy in nasopharyngeal carcinoma stem cells.Methods Nasopharyngeal carcinoma CNE-2 cells were selected to construct nasopharyngeal stem cell model,they were randomly divided into 0μmol/L group,4μmol/L group and 8μmol/L group by using random number table method.The proliferation of cells in each group was detected by CCK-8 cell proliferation test,Transwell cell migration test was used to detect the cell migration in each group,clonogenic assay was used to detect cell radiotherapy resistance,Western blot method was used to detect the expression of nuclear factor-k-gene-binding(NF-κB)p65 and hypoxia inducible factor-1α(HIF-1α)protein.Results OD value of cells in 4μmol/L group and 8μmol/L group was significantly lower than that in 0μmol/L group(P<0.05),and that in 8μmol/L group was significantly lower than that in 4μmol/L group(P<0.05).The number of perforating cells in 4μmol/L group and 8μmol/L group was significantly less than that in 0μmol/L group(P<0.05);the number of perforating cells in 8μmol/L group was significantly less than that in 4μmol/L group(P<0.05).The colony-forming ability of cells in 4μmol/L group and 8μmol/L group after 0 and 8 Gy irradiation was significantly lower than that in 0μmol/L group(P<0.05);the colony-forming ability of cells in 8μmol/L group after 0 and 8 Gy irradiation was significantly lower than that in 4μmol/L group(P<0.05).The expression of NF-κB p65 and HIF-1αin 4μmol/L group and 8μmol/L group was significantly lower than that in 0μmol/L group(P<0.05);the expression of NF-κB p65 and HIF-1αin 8μmol/L group was significantly lower than that in 4μmol/L group(P<0.05).Conclusion 2-ME2 can inhibit the proliferation and migration of nasopharyngeal carcinoma stem cells,and reduce the radiation resistance,the mechanism may be related to the inhibition of HIF-1/NF-κB signal transduction pathway,it is worthy of further study.
