摘  要:目的研究包载表阿霉素的乙酰普鲁兰叶酸偶合体纳米粒子(FPA/EPI)体内抗宫颈癌的作用及其机制。方法以人宫颈癌HeLa细胞移植瘤裸鼠为模型,分析尾静脉注射5 mg·kg^-1 FPA/EPI对荷瘤裸鼠体重、血液学、移植瘤体积和重量的影响,采用苏木精-伊红染色(HE)和原位末端标记法(TUNEL)观察FPA/EPI对移植瘤细胞凋亡的影响。结果FPA/EPI对HeLa细胞移植瘤裸鼠的体重未见明显影响,对肿瘤的生长具有抑制作用,肿瘤重量抑制率为32.37%。EPI及FPA/EPI均可诱导HeLa细胞凋亡。与对照组相比,FPA/EPI组网织红细胞显著降低(P<0.01)。结论FPA/EPI具有抗宫颈癌细胞移植瘤作用,但其药效和安全性尚需进一步的研究以明确。Objective To investigate the anti-cervical cancer effect and mechanism of epirubicin(EPI)loaded folate-targeted pullulan acetate(FPA)nanoparticles.Methods Based on the HeLa cells transplantation tumor model,the effects of 5 mg·kg^-1 FPA/EPI were investigated on the bodyweight,haematological variables,growth and weight of tumor tissues of the nude mice.The hematoxylin-eosin staining(HE)and terminal-deoxynucleotidyltransferase mediated nick end labeling method(TUNEL)were applied to observe the apoptosis of HeLa cells in the transplantation tumors.Results FPA/EPI showed no obvious adverse effects on the bodyweight of tumor-bearing nude mice.The tumor weight of FPA/EPI group was much less than that of control group,and the inhibition rate was 32.37%.EPI and FPA/EPI both could induce apoptosis of HeLa cells.Compared with control group,the reticulocyte of FPA/EPI group were reduced significantly(P<0.01).Conclusion FPA/EPI showed anticancer effects,but its efficacy and safety still need further research.
