摘  要:目的观察胃复方含药血浆对人胃癌BGC-823细胞凋亡及PPARγ、PTEN表达的影响,并探讨其作用机制。方法通过灌胃给药制备胃复方含药血浆,采用MTS法检测胃复方含药血浆对人胃癌BGC-823细胞生长的影响,并确定后续给药浓度。将体外培养的人胃癌BGC-823细胞随机分为胃复方含药血浆组(15%胃复方含药血浆)、空白对照组(15%磷酸缓冲盐溶液)、阳性对照组(15%氟尿嘧啶含药血浆)、阴性对照组(15%蒸馏水血浆),处理72 h后采用流式细胞术检测细胞凋亡,采用RT-PCR法、蛋白质印迹法分别检测PPARγ、PTEN mRNA或蛋白的表达水平。结果与空白对照组比较,胃复方含药血浆组与阳性对照组的细胞凋亡率、PPARγmRNA及蛋白表达水平、PTEN mRNA及蛋白表达水平均显著升高(P<0.05);与阴性对照组比较,胃复方含药血浆组与阳性对照组的细胞凋亡率、PPARγmRNA及蛋白表达水平、PTEN mRNA及蛋白表达水平均显著升高(P<0.05);但胃复方含药血浆组的细胞凋亡率、PPARγmRNA及蛋白表达水平、PTEN mRNA及蛋白表达水平均显著低于阳性对照组(P<0.05)。结论胃复方含药血浆可诱导人胃癌BGC-823细胞凋亡,其作用机制可能与活化的PPARγ上调PTEN表达有关。Objective To observe the effect of Weifufang-medicated plasma on apoptosis of human gastric cancer BGC-823 cells and expressions of PPARγ and PTEN, and to explore the mechanism of its effect on gastric cancer. Methods Weifufang-medicated plasma was prepared through gavage. MTS was used to detect the effect of Weifufang-medicated plasma on the growth of human gastric cancer BGC-823 cells, and the concentration of subsequent experiments was determined. BGC-823 cells cultured in vitro were randomly divided into three groups: Weifufang-medicated plasma group(15% drug containing plasma), blank control group(15% phosphate buffer salt solution), positive control group(15% fluorouracil containing plasma), and negative control group(15% distilled water containing plasma). After 72 hours of treatment, the apoptosis of BGC-823 cells was detected by flow cytometry, and the mRNA or protein expressions of PPARγ and PTEN were detected by RT-PCR assay or Western blotting. Results Compared with the blank control group, the apoptotic rate of BGC-823 cells, the expressions of PPARγ mRNA and protein, PTEN mRNA and protein were increased in the Weifufang-medicated plasma group and the positive control group(P<0.05). Compared with the negative control group, the apoptotic rate of BGC-823 cells, expressions of PPARγ and PTEN in both mRNA and protein level were increased in the Weifufang-medicated plasma group and the positive control group(P<0.05). However, the apoptotic rate of BGC-823 cells, and both mRNA and protein expression levels of PPARγ and PTEN were lower in Weifufang-medicated plasma group than in positive control group(P<0.05). Conclusion Weifufang-medicated plasma can induce apoptosis of human gastric cancer BGC-823 cells, and its mechanism may be related to the up-regulation of PTEN expression by activated PPAR γ.
