摘  要:本研究采用Illumina高通量测序技术对运动干预阻塞性黄疸致肝损伤模型小鼠的血液和肝组织进行测序,并验证运动在干预血液和肝组织炎症相关的因子差异表达的作用。本研究用雄性健康昆明种小鼠30只,随机分成假手术组、模型组和运动组,采用腹壁悬挂胆总管方法进行阻塞性黄疸致肝损伤造模。造模成功炎性期后,对运动组小鼠采取中等强度的跑台运动进行训练。对小鼠的血液和肝组织分别进行Illumina高通量测序,并对炎性相关差异表达因子进行相关性分析。结果显示,运动能有效地干预阻塞性黄疸致肝损伤所诱发的炎性反应,使炎症相关因子差异表达显著下降,与炎症和免疫相关的信号通路的富集表现显著降低。运动干预小鼠的血液和肝组织中差异表达显著的炎性相关的因子主要富集在NF-κB、TGF-β、TNF等信号通路的相关因子,其相关分子功能主要涉及到细胞信号转导、抑制细胞凋亡、免疫反应等。从KEGG富集的信号通路来看,小鼠血液和肝组织中的差异性显著富集通路也主要集中于NF-κB、TGF-β、TNF炎性相关的信号通路。且各炎性因子及炎性信号通路的差异表达在小鼠血液和肝组织内呈现相同趋势。结果表明中等强度的有氧运动能对阻塞性黄疸致肝损伤起到有效的抗炎效果,其表现在相关的炎性因子表达显著下降,且炎性信号通路的表达也显著降低,同时,各炎性因子和炎性信号通路的富集与差异表达呈现相同趋势。本研究为临床上采用有氧运动干预阻塞性黄疸致肝损伤患者的效果评定及其靶标因子筛选等提供实验基础。In this study,Illumina high-throughput sequencing technique was used to sequence the blood and liver tissue of mice with hepatic injury induced by obstructive jaundice,we verified the effect of exercise on the differential expression of inflammation related to blood and liver tissue.30 male rats were randomly divided into three groups:sham operation group,model group and exercise group.Using the method of hanging the common bile duct in the abdominal wall to establish the liver injury model induced by obstructive jaundice.After the successful establishment of the inflammations model,the mice in the exercise group were trained with moderate intensity treadmill exercise.Illumina high-throughput sequencing was performed on the blood and liver tissues of mice,and the correlation analysis of inflammatory-related differential expression factors was performed.The results showed that the exercise could effectively interfere with the inflammatory response induced by hepatic injury induced by obstructive jaundice,and the difference of inflammatory related factors was significantly decreased,and the enrichment performance of the signal pathways associated with inflammation and immunity was significantly reduced.The differentially expressed inflammatory related factors in the blood and liver tissues of the mice interfered with exercise were mainly enriched in NF-κB,TGF-β,TNF and other signaling pathways.The related molecular function mainly involved cell signal Transduction,inhibition of apoptosis,immune response.From the KEGG-enriched signaling pathway,the differences in blood and liver tissue of mice were mainly focused on inflammatory-related signaling pathways such as NF-κB,TGF-βand TNF.And the differential expression of inflammatory factors and inflammatory signaling pathways showed the same trend in mouse blood and liver tissue.The results showed that moderate aerobic exercise could play an effective anti-inflammatory effect on obstructive jaundice-induced liver injury,and its expression of related inflammatory fac
