摘  要:本研究利用PCR基因芯片,通过荧光定量PCR检测分析肿瘤坏死因子TNF-α对离体培养的大鼠下丘脑原代细胞不同神经递质受体的影响,以研究TNF-α对神经内分泌系统的影响。研究结果,发现后叶加压素1A受体(Avpr1a)、γ-氨基丁酸受体1 (Gabrr1)、离子型谷氨酸受体1 (Grin1)、5-羟色胺受体4 (Htr4)和转位蛋白(Tspo) 5个基因的表达有显著提高。另外肾上腺素能β2受体(Adrb2)、胆囊收缩素B受体(Cckbr)、毒蕈碱亚型胆碱能受体5 (Chrm5)、烟碱亚型胆碱能受体6 (Chrna6)、γ-氨基丁酸受体5,6 (Gabra5, Gabra6)、代谢型谷氨酸受体8 (Grm8)、5-羟色胺受体2c,3a (Htr2c, Htr3a)、生长抑素受体1 (Sstr1)和速激肽受体1(Tacr1) 11个基因的表达有明显下降。其中最为明显的是对离子型谷氨酸受体1 (Grin1)和速激肽受体1(Tacr1)的影响,分别是提高了3.12和降低了2.76。由此可见,TNF-α可以通过特定的信号传导途径调节下丘脑细胞中多种神经递质受体的转录表达水平,以改变神经元细胞对相应递质分子的应答水平和兴奋性,从而影响下丘脑组织内多种内分泌激素的释放量,并最终影响大鼠的神经内分泌功能。In this study, PCR gene chip and realtime fluorescence quantitative PCR were used to analyze the effect of tumor necrosis factor TNF-α on different neurotransmitter receptors in isolated cultured hypothalamic primary cells of rat in order to study the effect of TNF-α to neuroendocrine system. The result showed that the expression level of Avpr1 a, Gabrr1, Grin1, Htr4 and Tspo 5 genes was obviously up, the other Adrb2, Cckbr,Chrm5, Chrna6, Gabra5, Gabra6, Grm8, Htr2 c, Htr3 a, Sstr1 and Tacr1 11 genes were down. Especially, Grin1 and Tacr1 were affected obviously, increased 3.12 and decreased 2.76 respectively. So, TNF-α can modulate the expression of many receptors of neurotransmitters of hypothalamus by the special cell signal transduction pathways, and alter the responsibility and exciting of these neurons to neurotransmitters corresponding.Furthermore these changes can affect many hormones secretion of hypothalamus and effect neuroendocrine function of rat.
