摘 要:以竹叶兰新鲜花粉为试验材料,采用液体培养基,在单因素试验的基础上,设计正交试验,比较蔗糖、H 3BO 3和CaCl 2对竹叶兰花粉萌发的影响;另外,进行竹叶兰花粉贮藏性试验,测定竹叶兰单花花期内花粉萌发率的变化,旨在筛选出竹叶兰花粉离体萌发的适宜液体培养基组分配比和贮藏条件,为其种质资源保存等研究奠定基础。结果表明:(1)单因素试验中,在一定的浓度范围内的蔗糖、H 3BO 3和CaCl 2,对竹叶兰的花粉萌发具有一定的促进作用,超过一定浓度则起抑制作用;(2)正交试验中,蔗糖和CaCl 2对竹叶兰花粉萌发有显著影响,适宜的花粉离体萌发培养液为100 g·L^-1蔗糖+20 mg·L^-1H 3BO 3+5 mg·L^-1 CaCl 2,花粉萌发率58.71%;(3)贮藏性试验中,竹叶兰花粉在4、-7和-25℃中贮藏相同时间,温度越低,花粉活力相对保存越好;(4)竹叶兰单花花期约为3 d,开花当天花粉萌发率58.71%,而后花粉萌发率呈下降趋势。With fresh pollen of Arundina graminifolia as experimental material and adopting liquid culture medium,comparisons were made among the effects of sucrose,H 3BO 3 and CaCl 2 on the germination of A.graminifolia pollen by an orthogonal design based on the single factor tests,which was also used to investigate the pollen storage capacity.The aim of study was to screen out the ideal liquid medium composition and storage conditions for A.graminifolia,which would lay the foundation for the research of its genetic resources preservation.The results showed that:(1)in the single factor tests,pollen germination were promoted within a certain concentration range of sucrose,H 3BO 3 and CaCl 2;(2)from the orthogonal tests,sucrose and CaCl 2 significantly influence the pollen germination of A.graminifolia,the optimal culture medium was 100 g·L^-1 sucrose+20 mg·L^-1 H 3BO 3+5 mg·L^-1 CaCl 2,in which the rate of pollen germination was 58.71%;(3)in storage tests,the temperature of 4,-7,and-25℃were designed to store the pollen of A.graminifolia,at the same storage time,the lower temperature,the better preservation of pollen vigor;(4)the pollen vigor was the highest on the flowering day,in which the germination rate was 58.71%,and the germination rate decreased during the single flower period.
