摘 要:目的探讨骨髓基质干细胞(BMSCs)对脑缺血大鼠神经组织及功能的影响。方法选用24只SD大鼠制备大脑中动脉阻塞(MCAO)模型,随机分为移植组、注射组、对照组,每组8只。局部给予移植组动物BMSCs,由尾静脉给予注射组动物BMSCs,对照组动物尾静脉注射等量生理盐水。采用5-溴脱氧尿苷嘧啶(BrdU)标记新生细胞。MCAO术后第7、14天,每组各处死4只动物,采用改良神经功能缺损评分(mNSS)评估大鼠神经功能(处死前),取脑组织进行双标免疫组织化学染色,观察神经元样细胞和胶质细胞的生成情况。结果MCAO术后第7、14天,移植组和注射组动物BrdU+NES双标阳性细胞均多于对照组,差异有统计学意义(均P<0.05)。移植组动物术后第7、14天及注射组动物术后第7天BrdU+GFAP双标阳性细胞均少于对照组(均P<0.05)。术后第14天,移植组和注射组大鼠mNSS评分均低于对照组,差异有统计学意义(均P<0.05)。结论局部移植或静脉给予MCAO后大鼠BMSCs,可减少胶质瘢痕的形成,增加神经元样细胞的生成,改善神经功能。Objective To investigate the effects of bone marrow stromal cells(BMSCs)on the nervous tissue and function of cerebral ischemia rats.Methods Twenty-four SD rats were used to prepare the middle cerebral artery occlusion(MCAO)model.They were randomly divided into transplantation group,injection group and control group,with 8 rats in each group.BMSCs were locally administered in transplantation group and were administered from the tail vein in injection group.The same amount of normal saline was injected from the tail vein in control group.Neonatal cells were labeled with bromodeoxyuridine(BrdU).On the 7th and 14th day after MCAO,4 rats were sacrificed in each group.The modified neurological severity score(mNSS)was used to evaluate the neurological function of the rats(before sacrifice).Brain tissue was taken for double-label immunohistochemical staining to observe the formation of neuron-like cells and glial cells.Results On the 7th and 14th day after MCAO,BrdU+NES double-labeled positive cells in transplantation group and injection group were more than those in control group(all P<0.05).The number of BrdU+GFAP double-labeled positive cells on the 7th and 14th day after MCAO in transplantation group and the 7th day after MCAO in injection group were lower than those in control group(all P<0.05).On the 14th day after MCAO,the mNSS scores of rats in transplantation group and injection group were lower than those in control group,and the differences were statistically significant(both P<0.05).Conclusions Local transplantation or intravenous administration of BMSCs in cerebral ischemia rats could reduce the formation of glial scars,increase the production of neuron-like cells and improve neurological function.
