摘 要:目的评价将抗菌十肽KSL整合至牙本质粘接剂中,对其抗菌性能和粘接性能的影响。方法测定KSL对变异链球菌的最小杀菌浓度(MBC),按此浓度配制含KSL的粘接剂,制成实验组试件,对照组不加KSL,余处理同实验组。进行接触抑菌实验,计数两组粘接剂表面变异链球菌数和抑菌率。于试件表面接种人全菌生物膜接种液,利用扫描电子显微镜观察两组粘接剂表面生物膜形成情况。进行抗剪切力实验,观察两组粘接剂的粘接强度。结果抗菌十肽对变异链球菌的MBC值为125μg/mL。对照组和实验组粘接剂表面变异链球菌数分别为(4.80±0.93)×10^8、(2.96±0.64)×10^7 CFU/mL,差异有统计学意义(P<0.05),抑菌率为93.84%。电镜下可见实验组粘接剂表面细菌皱缩、裂解,无生物膜形成;对照组粘接剂表面有大量细菌,生物膜形成。对照组和实验组剪切力试验断裂强度分别为(258.92±13.69)、(250.17±25.65)N,差异无统计学意义(P>0.05)。结论将MBC浓度的抗菌十肽KSL整合至粘接剂中,可有效抑制变异链球菌的繁殖及生物膜的形成,不影响粘接剂强度。Objective To evaluate the effects of antibacterial decapeptide KSL on antibacterial and adhesive properties of dentin adhesives.Methods The minimum bactericidal concentration(MBC)of KSL against streptococcus mutans was determined.The KSL-containing adhesive was prepared at this concentration to make the test-pieces in experimental group.The test-pieces in control group was not added with KSL,and other operations were the same with experimental group.The contact bacteriostasis experiment was carried out,and the number of streptococcus of the two groups of adhesives and the bacteriostasis rate were counted.The whole-bacterium inoculum was inoculated on the surface of the test-pieces,and the formation of biofilm was observed by scanning electron microscope.A shear resistance test was conducted to observe the adhesive strength of the two groups of adhesives.Results The MBC of KSL against streptococcus mutans was 125μg/mL.The number of streptococcus mutans on the surface of adhesive in control group and experimental group were(4.80±0.93)×10^8 CFU/mL and(2.96±0.64)×10^7 CFU/mL,respectively.The difference was statistically significant(P<0.05).The inhibition rate was 93.84%.Under the electron microscope,the bacteria on the surface of the adhesive in experimental group shrank and cracked,and no biofilm was formed.On the surface of the adhesive in control group,a large amount of bacteria and biofilm formed.The adhesive strengths of control group and experimental group were(258.92±13.69)N and(250.17±25.65)N,respectively,and the difference was not statistically significant(P>0.05).Conclusions The integration of antibacterial decapeptide KSL into dentin adhesive could effectively inhibit the growth of streptococcus mutans and the formation of biofilm without affecting the strength of the adhesive.
