摘  要:目的:探讨分化抑制因子1(ID1)在结直肠癌肝转移中的意义及其作用机制。方法:收集2012年8月至2017年9月汕头大学医学院第一附属医院结直肠腺癌患者临床样本共50例,通过免疫组化方法检测ID1在发生肝转移的结直肠癌组织与未发生肝转移的结直肠癌组织的表达差异;通过过表达手段,检测ID1对RKO细胞上皮-间质转化的相关标志物的影响。结果:与癌旁正常组织相比,结直肠癌组织中ID1表达上调;与没有发生肝转移的癌组织相比,发生肝转移的癌组织中ID1表达上调,差异均具有统计学意义(均P<0.01)。在人结肠癌细胞系RKO中过表达ID1后,Twist和磷酸化的STAT3表达上调。结论:ID1可能是结直肠癌中潜在的促癌基因,ID1激活STAT3信号通路,引起Twist转录增强,可能与结直肠癌的肝转移相关。Objective:To investigate the role of inhibitor of differentiation 1(ID1)in liver metastasis of colorectal cancer and its mechanism.Methods:A total of 50 patients with colorectal adenocarcinoma from August 2012 to September 2017 in the First Affiliated Hospital of Medical College of Shantou University were collected.The expression of ID1 in colorectal cancer tissues with hepatic metastasis and colorectal cancer tissues without hepatic metastasis were detected by immunohistochemical method.The effect of ID1 on epithelial-mesenchymal transition markers in RKO cells was detected by overexpression.Results:The expression of ID1 in colorectal cancer tissues was up-regulated compared with that in adjacent normal tissues,and the expression of ID1 in hepatic metastasis tissues was up-regulated compared with that in non-hepatic metastasis tissues(all P<0.01).After overexpression of ID1 in human colon cancer cell line RKO,the expressions of Twist and phosphorylated STAT3 were up-regulated.Conclusion:ID1 may be a potential oncogene in colorectal cancer.ID1 activates STAT3 signaling pathway and induces enhanced Twist transcription,which may be related to liver metastasis of colorectal cancer.
