摘 要:目的:通过对类风湿关节炎(RA)患者外周血单个核细胞(PBMC)中核因子κB(NF-κB)、细胞型Fas相关死亡域样白介素-1β转换酶抑制蛋白(c-FLIP)的基因表达分析,探讨其mRNA表达水平在RA不同活动期的相关性。方法:选择2014年1月至2015年6月福建医科大学附属漳州市医院RA住院患者60例,按照28个关节疾病活动度评分进行分组,其中低活动期组22例、中活动期组20例、高活动期组18例。选择同期健康体检者25例作为健康对照组。采用荧光定量PCR检测RA患者和健康体检者PBMC中NF-κB、c-FLIP mRNA表达水平;对NF-κB与c-FLIP mRNA的表达进行线性回归相关性分析。结果:低、中活动期组NF-κB mRNA表达水平均高于健康对照组(均P<0.05),高活动期组NF-κB mRNA表达水平低于健康对照组(P<0.05)。低、中、高活动期组c-FLIP mRNA表达水平均高于健康对照组(均P<0.05)。NF-κB与c-FLIP的mRNA表达水平呈负相关(r=?0.495,P=0.021)。结论:RA患者PMBC中,c-FLIP mRNA表达水平随着RA病程的发展逐渐升高,与NF-κB mRNA表达水平负相关,可为RA患者NF-κB的靶向诊治研究提供参考。Objective:By analyzing the gene expression of nuclear factor-κB(NF-κB)and Fas-related death domain-like interleukin-1 beta-converting enzyme inhibitor protein(c-FLIP)in peripheral blood mononuclear cells(PBMC)of patients with rheumatoid arthritis(RA),the correlation of the expression level of their mRNA at different active stages of RA was explored.Methods:Sixty patients with RA in Zhangzhou Hospital Affiliated to Fujian Medical University from January 2014 to June 2015 were selected and grouped according to 28 joint disease activity scores.Among them,22 patients were in low activity stage,20 patients in middle activity stage and 18 patients in high activity stage.Twenty-five health examinees were selected as healthy control group during the same period.The mRNA expression levels of NF-κB and c-FLIP in PBMC of RA patients and healthy subjects were detected by fluorescence quantitative PCR,and the correlation between the mRNA expression of NF-κB and c-FLIP was analyzed by linear regression.Results:The mRNA expression level of NF-κB in low and middle active group was higher than that in healthy control group(all P<0.05),while the mRNA expression level of NF-κB in high active group was lower than that in healthy control group(P<0.05).The mRNA expression level of c-FLIP in low,middle and high activity group was higher than that in healthy control group(all P<0.05).The mRNA expression level of NF-κB was negatively correlated with that of c-FLIP(r=0.495,P=0.021).Conclusion:In PMBC of RA patients,the expression level of c-FLIP mRNA gradually increased with the development of RA,and was negatively correlated with the expression level of NF-κB,which could provide reference for the targeted diagnosis and treatment of NF-κB in RA patients.
