摘  要:目的:探讨miR-9对人牙周膜干细胞(PDLSCs)增殖、迁移和成骨能力的调控作用。方法:分离和培养人PDLSCs,瞬时转染miR-9 inhibitor片段后,使用Brdu法、细胞划痕实验、茜素红染色和Western blot法分别检测PDLSCs的增殖、迁移和成骨能力的改变。结果:瞬时转染miR-9 inhibitor后,PDLSCs的增殖能力减弱,细胞迁移速度变慢,成骨能力受损(P<0.05)。结论:miR-9可调控PDLSCs的增殖、迁移和成骨能力。Objective:To observe the effects of miR-9 on the proliferation,migration and osteogenic differentiation of human periodontal ligament stem cells(PDLSCs)in vitro.Methods:PDLSCs were isolated and cultured in vitro,and were transfected with miR-9 inhibitor.Proliferation of PDLSCs was evaluated by Brdu assay.Migration of PDLSCs was tested by cell wound scratch assay.Osteogenic differentiation of PDLSCs was determined by Alizarin red staining and Western blot.Results:Suppression of the function of miR-9 could reduce proliferation of PDLSCs,lowered the migration speed of PDLSCs,and harmed the osteogenic differentiation of PDLSCs(P<0.05).Conclusions:miR-9 could regulate the proliferation,cell migration and osteogenic differentiation of PDLSCs.
