摘  要:目的:体外构建釉质矿化模型,在脱矿的釉质表面重建类釉质晶体结构。方法:将特制的Y型三通管底端封闭,从上端加入含有CaCl2的琼脂凝胶,脱矿的釉质片放在其表面,同时在牙片表面覆盖CaCl2琼脂和纯琼脂凝胶,再加入含或不含氟的Na2HPO4溶液,并连接阴极;三通管侧端加入CaCl2溶液,并连接阳极,施加40 mA直流电场。分别在不同的矿化时间,取出釉质片,进行场发射扫描电镜观察。结果:在不加电流情况下,不含氟离子时,在脱矿的釉质表面形成了少量且不均匀的条带状晶体;含有氟离子时,在脱矿的釉质表面形成了六边形的且层层密集堆积的片状晶体。在直流电场情况下,不含氟离子时,在釉质表面沉积的晶体量增多,晶体的形态也发生改变为片状晶体;在含有氟离子时,脱矿釉质表面形成了棒状的晶体且相互平行排列,类似釉柱结构。结论:外加电场的载钙磷氟离子的琼脂凝胶矿化体系可以仿生釉质矿化的过程,可使脱矿的釉质表面沉积类似釉柱样的晶体结构。Objective:Regenerating enamel-like mineralized tissue on the demineralized enamel surface by constructing an enamel mineralization model in vitro.Methods:The bottom end of the three-way tube is closed,and CaCl2 agarose gel was placed in the tube to separate the upper and side end,and acid-etched enamel slice was placed on the gel surface from upper end.A 2 mm thick layer of agarose gel containing CaCl2 was used to cover acid-etched enamel slice,followed by a 2 mm thick layer of ion-free agarose gel.Then Na2HPO4 solution(or containing sodium fluoride)was added onto the ion-free gel surface.CaCl2 solution was added to the side tube and was connected to the anode.Na2HPO4 solution was connected to the cathode,and 40 mA DC electric field was applied.Results:When no current was applied,a small amount of uneven banded crystals formed on the demineralized enamel surface without fluoride ions;Hexagonal and densely packed flake crystals formed on the demineralized enamel surface with fluoride ions.In the case of a DC electric field,with no fluorine ions,the amount of crystal deposited on the enamel surface increases,and the morphology of the crystal changes to a plate crystal.With fluoride ion,the surface of the demineralized enamel forms a nanorod-like crystal and is parallel to each other which was similar to enamel prisms.Conclusions:The agarose gel mineralization system loaded with calcium phosphate and fluoride ions aided with an electric field can biomimetic the process of enamel mineralization,which can deposit an enamel prism-like crystal structure on the demineralized enamel surface.
