摘 要:目的研究姜黄素对类风湿关节炎(RA)破骨细胞(OC)分化中核因子κB受体活化因子(RANK)基因及蛋白表达的影响。方法采用核因子κB受体活化因子配体(RANKL)和巨噬细胞集落刺激因子(M?CSF)诱导RA病人外周血单个核细胞(PBMC)向OC分化,给予不同浓度的姜黄素(2.5、5及10μmol/L)进行干预,并设空白对照组。抗酒石酸酸性磷酸酶(TRAP)染色标记成熟OC并计数;实时荧光定量聚合酶链式反应(Real?time PCR)和蛋白质印迹法(Western Blot)检测各组细胞RANK基因表达和蛋白水平。结果RA病人PBMC经RANKL诱导后能获得大量TRAP阳性的OC,经姜黄素(2.5、5及10μmol/L)干预后,TRAP阳性细胞数明显减少(P<0.05);与空白对照组相比,姜黄素各浓度组细胞RANK基因表达显著降低(P<0.05);姜黄素(2.5、5及10μmol/L)各浓度组细胞RANK蛋白的表达分别为(0.46±0.09)、(0.36±0.08)和(0.25±0.07),与空白对照组(0.68±0.11)相比均显著降低(F=21.278,P=0.000)。结论姜黄素可能通过下调RANK基因和相关蛋白的表达,抑制RA病人OC分化。Objective To study the effect of Curcumin on the expression of receptor activator for nuclear factor?κB(RANK)gene and protein in osteoclast(OC)differentiation of rheumatoid arthritis(RA).Methods RANK ligand(RANKL)and macrophage colony?stimulating factor(M?CSF)were used to induce peripheral blood mononuclear cells(PBMCs)from patients with RA to dif?ferentiate into OC.During this process,different concentrations of Curcumin(2.5,5,and 10μmol/L)were given to intervention and a blank control group was set up.The mature OCs were labeled with tartrate?resistant acid phosphatase(TRAP)and counted.Real?time PCR and Western blot were used to detect the RANK gene expression and protein levels in each group.Results A large num?ber of TRAP?positive OCs was obtained from PBMCs of RA patients induced by RANKL.After intervention with Curcumin(2.5,5,and 10μmol/L),the number of TRAP?positive cells was significantly reduced(P<0.05).Compared with the blank control group,the expression of RANK gene in the cells of each concentration group of Curcumin was significantly decreased(P<0.05).The ex?pressions of RANK protein in Curcumin(2.5,5 and 10μmol/L)groups were(0.46±0.09),(0.36±0.08)and(0.25±0.07),respec?tively,which were significantly lower than(0.68±0.11)of the blank control group(F=21.278,P=0.000).Conclusion Curcumin inhibits OC differentiation in RA patients by down?regulating the expression of RANK gene and related protein.
