Artemin CXCL13 MMP-9在子宫内膜癌组织中的表达及临床意义

摘　　要：目的:研究Artemin、CXCL13和MMP-9在子宫内膜癌组、非典型增生组、单纯性增生组和正常内膜组,以及在不同临床病理特征内膜癌组织中的表达,分析其差异性表达与子宫内膜癌临床病理分期的关系。方法:选取承德医学院附属医院妇科2016年6月至2018年4月手术切除子宫内膜标本共154例,其中子宫内膜癌Ⅰ期56例(EC-Ⅰ组),Ⅱ期15例(EC-Ⅱ组),Ⅲ期7例(EC-Ⅲ组),非典型增生32例(EAH组),单纯性增生46例(ESH组),正常内膜组织44例(NE组)。Western Blot法检测Artemin、CXCL13和MMP-9蛋白在不同内膜组织中的表达,实时荧光定量PCR技术检测Artemin、CXCL13、MMP-9 mRNA在各组子宫内膜癌组织中的表达,分析其表达情况与子宫内膜癌临床病理特征的关系。结果:Artemin、CXCL13、MMP-9蛋白在EC组的表达量高于EAH、ESH和NE组(F=38.54,F=98.74,F=72.46,P均<0.05)。Artemin、CXCL13、MMP-9 mRNA在子宫内膜癌中的表达与肌层浸润深度、病理分化程度、FIGO分期及淋巴结转移相关(P均<0.05),与患者年龄不相关(P>0.05)。结论:Artemin、CXCL13、MMP-9在子宫内膜癌的发生发展中发挥作用,可为子宫内膜癌的早期诊断及预后判断提供新的研究方向。Objective:To investigate the expression of Artemin,CXCL13 and MMP-9 in endometrial carcinoma,atypical hyperplasia,simple hyperplasia and normal endometrial tissue,and in different clinicopathological features of endometrial carcinoma.The relationship between differential expression and clinical pathological staging of endometrial carcinoma.Methods:A total of 154 cases of endometrial specimens were removed from the Department of Gynecology,Affiliated Hospital of Chengde Medical College from June 2016 to April 2018,including 78 cases of endometrial carcinoma,56 cases of stage I(EC-I group),15 cases of stage II(EC-II group),7 cases of stage III(EC-III group),32 cases of atypical proliferative endometrial tissue(EAH group),46 cases of simple proliferative endometrial tissue(ESH group),normal endometrial tissue in 44 cases(NE group).The expression of Artemin,CXCL13 and MMP-9 in endometrial carcinoma,atypical hyperplasia,simple hyperplasia and normal endometrium was detected by Western Blot.Real-time quantitative PCR was used to detect Artemin,CXCL13 and MMP-9 mRNA.The expression of different clinicopathological features in endometrial carcinoma tissues was analyzed,and the relationship between differential expression and clinical pathological staging of endometrial carcinoma was analyzed.Results::The expression of Artemin,CXCL13 and MMP-9 in the EC group were higher than those in the EAH group,ESH group and NE group(F=38.54,F=98.74,F=72.46,all P<0.05).The expression of Artemin,CXCL13 and MMP-9 mRNA in endometrial carcinoma was correlated with depth of myometrial invasion,pathological differentiation,FIGO stage and lymph node metastasis(all P<0.05),which was not correlated with age(P>0.05).Conclusion:Artemin,CXCL13 and MMP-9 play a role in the development of endometrial carcinoma,which may provide new research direction for early diagnosis and prognosis of endometrial carcinoma.
