摘  要:目的探讨LSP1 rs3817198基因位点多态性与乳腺癌的关系。方法采集2005年6月至2015年1月间130例乳腺癌患者(乳腺癌组)和205例健康女性(对照组)外周血提取DNA,对LSP1基因rs381719位点进行检测。分析LSP1 rs3817198位点基因型与乳腺癌易感性的关系,采用Logistic回归分析影响乳腺癌发病因素。结果两组基因型分布均符合Hardy-Weinberg遗传平衡定律。LSP1 rs3817198位点的CC、CC+TC基因型频率分布在乳腺癌组和对照组间差异有统计学意义(P=0.018,P=0.017)。携带C等位基因较T等位基因者增加了患乳腺癌的风险(OR=1.726,95%CI=1.456~6.172,P=0.003)。LSP1 rs3817198位点基因型与乳腺癌患者PR状态有关,LSP1 TC/CC基因携带者的PR阴性率(50.4%)显著高于TT携带者的PR阴性率(23%),差异具有统计学意义(P=0.041)。结论LSP1 rs3817198基因的遗传多态性可增加女性罹患乳腺癌的风险,与PR阴性高表达有关,LSP1基因型可作为检测妇女乳腺癌易感性的重要生物标志物。Objective To investigate the relationship between LSP1 rs3817198 gene polymorphism and breast cancer.Methods DNA was extracted from peripheral blood of 130 breast cancer patients and 205 healthy women from June 2005 to January 2015,and the rs3817198 locus of LSP1 gene was detected.The relationship between LSP1 rs3817198 genotype and susceptibility to breast cancer was analyzed,and the influencing factors of breast cancer were analyzed by logistic regression.Results The distribution of genotypes in both groups conformed to Hardy-Weinberg s law of genetic balance.The frequencies of C and CC+TC genotypes at LSP1 rs3817198 locus were significantly different between breast cancer group and control group(P=0.018,P=0.017).Carriers of C allele had a higher risk of breast cancer than those of T allele(OR=1.726,95%CI=1.456-6.172,P=0.003).The genotype of LSP1 rs3817198 was related to PR status in breast cancer patients.The PR negative rate of LSP1 TC/CC carriers(50.4%)was significantly higher than that of TT carriers(23%).The difference was statistically significant(P=0.041).Conclusion The genetic polymorphism of LSP1 rs3817198 gene can increase the risk of breast cancer in women,which is related to the high expression of PR negative.LSP1 genotype can be used as an important biomarker to detect the susceptibility of breast cancer in women.
