摘 要:为分析猪源抗菌肽NK-lysin的理化性质和蛋白结构,并研究猪源抗菌肽在小鼠体内的抑菌效果,以猪小肠组织cDNA为模板进行PCR扩增和测序,通过生物信息学相关软件和工具对其测序结果进行分析。试验选用60只小鼠,随机分为4组(预防组、细菌模型组、治疗组和对照组),设置不同的大肠杆菌和猪源抗菌肽给药方案。结果表明,猪源抗菌肽NK-lysin为稳定的,半衰期在大肠杆菌中(体内)长,与白细胞介素-2(Interleukin-2,IL-2)和颗粒性蛋白B(Granzyme B,GZMB)具有相互作用;预防组和治疗组小鼠存活率分别为80.00%和86.67%。综上所述,猪源抗菌肽在小鼠体内安全且具有一定的抑菌作用。Physicochemical properties and protein structure of porcine antibacterial peptide NK-lysin were analyzed,and the antibacterial effect of porcine antibacterial peptides in mice was studied in the paper.The PCR amplification and sequencing were performed using porcine small intestine tissue cDNA as a template.Sequencing results were analyzed by bioinformatics related software and tools.Sixty mice were randomly divided into 4 groups according to test requirements,and 15 mice in each group were divided into a prevention group,a bacterial model group and a control group.Different treatments with porcine antibacterial peptide and E.coli were conducted.Results showed that porcine antibacterial peptide NK-lysin was stable,and its half-life was long in E.coli(in vivo),interacting with Interleukin-2(IL-2)and Granzyme B(GZMB).The survival rates of the prevention group and the treatment group were 80.00%and 86.67%,respectively.In conclusion,porcine antibacterial peptide was safe and had certain antibacterial activity in mice.
