基于TRB3基因调控探讨重楼皂苷Ⅰ抑制肝癌细胞增殖的研究

摘　　要：目的通过TRB3的表达研究重楼皂苷Ⅰ抑制肝癌细胞增殖的机制。方法CCK8法检测不同浓度的重楼皂苷Ⅰ作用不同时间对肝癌细胞HepG2和MHCC-97H增殖的影响;Western Blot和RT-qPCR检测重楼皂苷Ⅰ作用后,肝癌细胞HepG2和MHCC-97H的TRB3表达情况;shRNA及CCK8法检测TRB3基因对重楼皂苷Ⅰ抑制HepG2和MHCC-97H细胞增殖作用的影响。结果不同浓度的重楼皂苷Ⅰ作用不同时间均能够抑制肝癌细胞HepG2和MHCC-97H的增殖,这种抑制作用呈现浓度和时间依赖;Western Blot和RT-qPCR结果显示,在重楼皂苷Ⅰ作用下,肝癌细胞HepG2和MHCC-97H细胞中的TRB3蛋白和mRNA表达均显著增强(P<0.05);CCK8检测结果显示,与shRNA-TRB3组比较,重楼皂苷Ⅰ+shRNA-TRB3组肝癌细胞HepG2及MHCC-97H的存活力均显著增高(P<0.05)。结论重楼皂苷Ⅰ可通过增加肝癌细胞TRB3表达,抑制细胞增殖。Objective The mechanism of inhibition of hepatocellular carcinoma(HCC)cells proliferation by Polyphylla saponins I(PS I)was studied by the expression of TRB3.Methods CCK8 method was used to detect the effects of different concentrations of PS I on the proliferation of HCC cells HepG2 and MHCC-97H.Western Blot and RT-qPCR were used to detect the expression of TRB3 in HCC cells HepG2 and MHCC-97H.ShRNA and CCK8 assay were used to detect the effect of TRB3 gene on the proliferation of HepG2 and MHCC-97H cells by PS I.Results Different concentrations of PS I could inhibit the proliferation of HCC cells HepG2 and MHCC-97H at different times.The inhibition effect was concentration and time dependent.The expression of TRB3 protein and mRNA in HepG2 and MHCC-97H cells were significantly increased(P<0.05)under the influence of PS I.CCK8 assay showed that the viability of HCC cells HepG2 and MHCC-97H of PS I+shRNA-TRB3 group was significantly higher than that of shRNA-TRB3 group(P<0.05).Conclusion PS I can inhibit cell proliferation by increasing the expression of TRB3 in HCC cells.
