摘 要:目的研究脊柱结核患者Toll样受体-2(Toll like receptor 2,TLR-2)表达与血清中炎性因子及应激因子的相关性。方法收集2015年8月至2018年10月住院手术治疗的椎间盘突出症患者40例作为对照组,脊柱结核患者40例作为观察组。入院后采集两组患者静脉血,采用免疫组化检测TLR-2表达,采用Western blot检测TLR-2蛋白表达,采用Elisa法检测血清白细胞介素1β(Interleukin,IL-1β)、白细胞介素6(Interleukin,IL-6)和C反应蛋白(C reactive protein,CRP)含量,采用Person检验分析TLR-2与IL-1β、IL-6和CRP相关性。结果观察组TLR-2表达量(0.81±0.06)显著高于对照组TLR-2表达量(0.43±0.05)(t=13.447,P<0.05);观察组TLR-2蛋白相对表达量(0.63±0.05)显著高于对照组TLR-2蛋白相对表达量(0.37±0.07)(t=46.848,P<0.05);观察组血清IL-1β(77.23±11.22)pg/ml、IL-6(61.39±13.73)pg/ml和CRP(49.29±8.98)pg/ml显著高于对照组IL-1β(48.33±12.32)pg/ml、IL-6(32.63±9.32)pg/ml和CRP(29.89±14.37)pg/ml(P<0.05);TLR-2蛋白与血清IL-1β(r=0.762,P<0.05)、IL-6(r=0.689,P<0.05)和CRP(r=0.721,P<0.05)含量之间呈正相关。结论脊柱结核血清中IL-1β、IL-6和CRP可被认为与TLR-2有密切关系,有效抑制TLR-2可有效降低脊髓结核患者的炎症水平。Objective To investigate the correlation between toll like receptor 2(TLR-2)expression and inflammatory factors and stress factors in patients with spinal tuberculosis.Methods Data of 40 patients with disc herniation who were hospitalized from August 2015 to October 2018 were collected as control group and those of 40 patients with spinal tuberculosis as observation group.After admission,venous blood was collected.TLR-2 expression was detected by immunohistochemistry.TLR-2 protein expression was detected by Western blot,serum interleukin-β(IL-1β),IL-6 and C reactive protein(CRP)were detected by Elisa method.The correlation between TLR-2 and IL-1βand the correlation between IL-6 and CRP was respectively analyzed by Person test.Results The expression of TLR-2 in the observation group was significantly higher than that in the control group(0.81±0.06 v.s.0.43±0.05)(t=13.447,P<0.05).The relative expression of TLR-2 protein in the observation group was significantly higher than that in the control group(0.63±0.05 v.s.0.37±0.07)(t=46.848,P<0.05).The serum levels of IL-1β,IL-6 and CRP were(77.23±11.22)pg/ml,(61.39±13.73)pg/ml and(49.29±8.98)pg/ml in the observation group were significantly higher than those in the control group(48.33±12.32,32.63±9.32,29.89±14.37)(P<0.05).TLR-2 protein was positively correlated with serum levels of IL-1β (r=0.762, P< 0.05), IL-6 (r=0.689, P< 0.05) and CRP (r=0.721, P< 0.05). Conclusion SerumIL-1β, IL-6 and CRP in spinal tuberculosis can be considered to be closely related to TLR-2.Effective suppression of TLR-2 can reduce inflammation levels in patients with spinal tuberculosissignificantly.
