摘  要:目的探讨右美托咪定对大鼠局灶性缺血再灌注脑组织线粒体通透性转换孔(m PTP)的影响。方法成年雄性SD大鼠45只,随机分为Ⅰ组(空白对照组)、Ⅱ组(缺血再灌注组)、Ⅲ组(右美托咪定组)各15只。采用线栓法阻塞大脑中动脉,然后恢复灌注,制备大鼠脑缺血再灌注损伤模型(MCAO),各组于脑缺血2 h后进行再灌注,再灌注22 h后处死大鼠,取脑组织,采用TTC染色法测定脑梗死体积,采用分光光度法测定线粒体mPTP开放程度。结果Ⅱ组和Ⅲ组的脑梗死体积比和mPTP开放程度高于Ⅰ组(P<0.05);Ⅲ组的脑梗死体积比低于Ⅱ组(P<0.05),Ⅱ组和Ⅲ组的mPTP开放程度比较无统计学差异(P>0.05)。结论右美托咪定可减轻大鼠局灶性缺血再灌注损伤,但对mPTP开放程度无明显影响。Objective To study the influence of dexmedetomidine on mitochondrial permeability transition pore(m PTP)in brain tissues of rats with focal ischemia-reperfusion.Methods 45 male adult SD rats were randomly divided into groupⅠ(blank control group),groupⅡ(ischemia-reperfusion group)and groupⅢ(dexmedetomidine group),with 15 rats in each group.The suture embolization method was used to block the cerebral artery,and then the perfusion was recovered,so as to establish the middle cerebral artery occlusion(MCAO)model.All groups received perfusion 2 hours after cerebral ischemia,then the rats were killed 22 hours after perfusion for brain tissues.The cerebral infarct volume was detected by TTC,and the m PTP opening degree was detected by spectrophotometry.Results The cerebral infarct volume ratio and m PTP opening degree of groupⅡand groupⅢwere significantly higher than that of groupⅠ(P<0.05).The infarct volume ratio of groupⅢwas significantly lower than that of groupⅡ(P<0.05).No statistical difference was found in the m PTP opening degree between groupⅡand groupⅢ(P>0.05).Conclusions Dexmedetomidine can relieve the focal ischemia-reperfusion injury in rats,but has no significant influence on the m PTP opening degree.
