摘  要:热激蛋白70 (heat shock protein 70, Hsp70)是细胞应对高温和其它逆境胁迫时所产生的一类分子伴侣类型应激蛋白。本研究以向日葵(Helianthus annuus)基因组数据库为基础,利用生物信息学方法对其Hsp70基因家族成员进行鉴定和分析。结果表明,向日葵共有103个Hsp70基因家族成员,根据亚细胞定位结果可分为5种不同类型,分析了不同成员编码蛋白的理化性质。系统进化分析结果显示,HaHsp70基因家族成员分为9个分支,每个分支成员的结构和功能相对保守。根据MEME软件进行保守基序分析,并鉴定了103个基因家族成员在向日葵17条染色体上的数量及分布情况,其中以15号染色体上分布最多,而11号染色体没有成员分布。转录本结构分析展示了不同基因成员内含子以及外显子的结构情况。本研究结果有助于了解向日葵Hsp70基因家族成员的情况,进而为进一步研究向日葵Hsp70基因家族成员的功能分析提供基础。Heat shock proteins 70(heat shock protein 70, Hsp70) were induced under high temperature and other abiotic stress. In this study, 103 Ha Hsp70 gene family members were identified and analyzed from the reference genome of Helianthus annuus using bioinformatics. The results showed that there were five different types according to the subcellular location predication, and the molecular weight and isoelectric point of the protein were predicted. Phylogenetic analysis revealed that the Helianthus annuus Hsp70 gene family was categorized into nine major sub-families, in each sub-family, the constituent parts of the gene structure and motif were relatively conserved. MEME web server was employed as a method to analyze motif distribution, all identified HaHsp70 genes were mapped to 17 chromosomes of Helianthus annuus. Among them, chromosome 15 had the highest density of HaHsp70 genes, no Ha Hsp70 genes were detected in chromosome 11. The exon and intron organization of HaHsp70 was depicted using Gene Structure Display Server online. Overall, data obtained from our investigation contributes to a better understanding of the complexity of the Helianthus annuus Hsp70 gene family and provides the first step towards directing future experimentation designed to perform systematic analysis of the functions of the Hsp70 gene family.
