摘 要:目的:观察青光安缓释剂对抗青光眼术后滤过道瘢痕化过程中TGF-β/Smad信号传导通路相关蛋白的影响,探讨其作用机制。方法:将48只实验用兔分为空白组、模型组、MMC组、青光安组分1~4组和安慰剂组。除空白组外,其余各组行小梁切除+虹膜根部切除术,并进行相对应的处理。术后眼部常规检查,用药4周后进行术眼HE染色和相关蛋白检测。结果:术后MMC组和青光安组分2组眼压低于其他各组(P<0.05);两组滤过道轮廓清晰,纤维细胞及炎症细胞较其他各组少,且两组CollagenⅠ蛋白量明显低于其他各组(P<0.01);而MMC组Smad3、Smad4的蛋白量明显低于其他各组(P<0.01);结论:青光安组分2能使TGF-β/Smad通路的信号传导减弱,抑制滤过道瘢痕形成,安全性优于MMC。Objective: To observe the effect of Qingguangan slow-release agent on the related protein of TGF-β/Smad signal transduction pathway in the filtering scarring after glaucoma filtering surgery. To explore its mechanisms of Qingguangan slow release agent. Methods: Forty-eight rabbits were randomly divided into control group, model group, MMC group, Qingguanganan group component 1 to 4 group and the placebo group. In addition to the control group, trabeculectomy and irid root resection were performed in the other groups, and corresponding treatment was carried out. Taking routine examination of the eyes after operation, HE staining and related protein detection were performed 4 weeks later. Results: IOP in MMC group and Qingguangan component 2 group was lower than that of other groups(P<0.05). The outline of the filter passage in both groups was clear, and the number of fibroblasts and inflammatory cells in the two groups was less than that in the other groups. The protein content of Collagen Ⅰ in the two groups was significantly lower than that in the other groups(P<0.01). The protein content of Smad3 and Smad4 in MMC group was significantly lower than that in other groups(P<0.01). Conclusion: Qingguangan component 2 can weaken the signal transduction of TGF-β/Smad pathway and inhibit the formation of filtering scar, which is safer than MMC.
