摘  要:目的:观察肝豆扶木汤(GDFMT)对Wilson病肝纤维化毒乳鼠(TX)小鼠肝组织转化生长因子β受体1(TβRⅠ)、2(TβRⅡ)及Smad4的影响。方法:40只TX小鼠随机分为模型组,GDFMT高、中、低剂量组及青霉胺组,每组8只,8只DL小鼠作为正常组。各组给予相应药物,4周末检测小鼠血清透明质酸(HA)、层粘连蛋白(LN)、Ⅲ型前胶原蛋白(PC-Ⅲ)、Ⅳ型胶原蛋白(C-Ⅳ)含量,免疫组化和RT-PCR测定肝组织TβRⅠ、TβRⅡ及Smad4蛋白和mRNA表达。结果:与模型组比较,GDFMT高、中、低剂量组可以显著降低TX小鼠HA、LN、PC-Ⅲ、C-Ⅳ水平,改善肝纤维化病理,抑制TβRⅠ、TβRⅡ及Smad4蛋白和mRNA表达(P<0.01,P<0.05)。结论:GDFMT可以有效地逆转TX小鼠肝纤维化,其机制可能与抑制肝组织TβRⅠ、TβRⅡ及Smad4蛋白和mRNA表达有关。Objective: To observe the effects of Gandou Fumu Decoction on TβRⅠ, TβRⅡ and Smad4 expressions in TX mice liver with Wilson’s disease and live fibrosis. Methods: Forty TX mice were randomly divided into the model group, the penicillamine group, Gandou Fumu Decoction high, middle, low dose groups, 8 mice in each group, and 8 DL mice were used as control group. Each group was given corresponding drugs. After administration for four weeks, the levels of HA, LN, C-Ⅳ and PC-Ⅲ were determined. Liver samples were taken to examine the degree of liver fibrosis by masson staining. The protein and mRNA expression of TβRⅠ, TβRⅡ and Smad4 were detected by immunohisto chemistry and RT-PCR. Results: As compared with the model group, Gandou Fumu Decoction significantly reduced histopathological change, decreased the levels of HA, LN, PC-Ⅲ and C-IV. Meanwhile Gandou Fumu Decoction could effectively inhibit the protein and mRNA expression of TβRⅠ, TβRⅡ and Smad4 in TX mice liver(P<0.01, P<0.05). Conclusion: Gandou Fumu Decoction can resist Wilson disease liver fibrosis possibly by down-regulating the protein and mRNA expression TβRⅠ, TβRⅡ and Smad4.
