摘 要:目的 :探讨加味黄芪建中汤通过IL-33介导肿瘤相关巨噬细胞极化从而抑制肿瘤的研究。方法:采用多因素法建立小鼠脾虚证模型,完成后于皮下接种Lewis肺癌细胞悬液建立脾虚证荷瘤小鼠模型。使用加味黄芪建中汤干预治疗,检测小鼠血清IL-33水平,肿瘤组织中IL-33与巨噬细胞标记物F4/80的共表达及M1、M2型巨噬细胞标记物CD86、CD206的表达。结果:与脾虚肺癌组比较,黄芪建中汤组小鼠血清及肿瘤组织中IL-33表达显著降低(P<0.05),IL-33在巨噬细胞中的表达显著降低(P<0.05),肿瘤组织中CD206表达显著降低(P<0.01),CD86表达显著升高(P<0.05)。结论:加味黄芪建中汤治疗可改善脾虚状态,并通过下调IL-33的表达,使M2型向M1型巨噬细胞转化,从而达到抑制肿瘤转移的目的。Objective: To investigate the effects of Jiawei Huangqi Jianzhong Decoction on inhibiting tumor by polarization of tumor-related macrophages mediated by IL-33. Methods: The model of spleen deficiency syndrome in mice was established by multi-factor method. After completion, the model of spleen deficiency syndrome in mice was established by subcutaneous inoculation of Lewis lung cancer cell suspension. The levels of IL-33 in serum, co-expression of IL-33 with macrophages marker F4/80 and M1, M2 macrophages markers CD86 and CD206 in tumor tissues were detected with Jiawei Huangqi Jianzhong Decoction. Results: Compared with the spleen deficiency lung cancer group, the expression of IL-33 in serum and tumor tissue of the treated group was decreased(P<0.05);the expression of IL-33 in macrophages was decreased(P<0.05);the expression of CD206 was increased(P<0.01) and the expression of CD86 was decreased in the Huangqi Jianzhong Decoction group(P<0.05). Conclusion: Modified Huangqi Jianzhong Decoction can improve the state of spleen deficiency and inhibit the metastasis of tumors by down-regulating the expression of IL-33 and transforming M2 into M1 macrophages.
