摘 要:目的探讨壳寡糖对胰腺癌细胞系PACN-1生长的影响作用。方法将培养至对数期生长的PACN-1细胞根据加入壳寡糖的浓度将其分为壳寡糖1.0 mg/ml组、壳寡糖2.0 mg/ml组及空白组。通过细胞集落实验观察并记录各组细胞集落形成的特点。通过四唑盐比色法(MTT)实验,比较各组细胞的增殖情况。结果细胞集落实验发现,培养48 h、96 h后,随壳寡糖浓度的升高,细胞集落的细胞数减少,细胞集落的的细胞延展性变差,细胞间的连接程度变好。MTT实验发现,培养48 h后,壳寡糖对PACN-1生长有较显著的抑制作用,且随壳寡糖浓度的升高及时间的延长,其对胰腺癌细胞生长的抑制作用增强。结论壳寡糖在体外研究中有抑制PACN-1细胞生长的作用,可能为胰腺癌治疗提供了一种新的途径。Objective To investigate the effect of chitooligosaccharides on the growth of pancreatic cancer cell line PACN-1.Methods PACN-1 cells were cultured to logarithmic phase.Colony formation of PACN-1 cells in blank group and with different concentrations of chitooligosaccharides(1.0 and 2.0 mg/ml)were observed and recorded by cell colony assay.The proliferation of cells was compared by MTT assay in each group.Results It was found in the cell colony experiment that,after 48 hours and 96 hours of culture,the number of cells in the cell colony reduced with the increase of chitooligosaccharides concentration,the ductility of cells in the cell colony became worse,and the degree of connection between cells became better.MTT assay showed that chitooligosaccharides had a significant inhibitory effect on the growth of PACN-1 after 48 hours of culture,and its inhibitory effect on the growth of pancreatic cancer cells was enhanced with the increase of chitooligosaccharides concentration and the extension of time.Conclusion Chitooligosaccharides can inhibit the growth of PACN-1 cells in vitro,which may provide a new way for the treatment of pancreatic cancer.
