摘　　要：目的分析靶向沉默S100A4基因对子宫内膜癌细胞顺铂化疗敏感性的影响。方法病毒转染子宫内膜癌KLE细胞株并获得稳定细胞株;聚合酶链反应检测转染后子宫内膜癌细胞中S100A4 mRNA的表达;Western blot印迹法检测转染后子宫内膜癌细胞中S100A4蛋白的表达水平;CCK8法检测转染后子宫内膜癌细胞的增殖变化。结果阴性对照组、shRNA干扰实验组、过表达实验组3组之间光密度值两两比较,差异均有统计学意义(P<0.05)。阴性对照组、shRNA干扰实验组、过表达实验组的S100A4相对表达水平两两比较,差异均有统计学意义(P<0.05)。空白对照组、shRNA干扰实验组的S100A4蛋白表达水平明显低于阴性对照组,过表达实验组的S100A4蛋白表达水平明显高于阴性对照组,差异均有统计学意义(P<0.05)。空白对照组、阴性对照组、shRNA干扰实验组、过表达实验组的IC50值两两比较,差异均有统计学意义(P<0.05)。结论靶向沉默S100A4基因可抑制肿瘤细胞增殖,促进细胞凋亡,增强子宫内膜癌细胞对顺铂的敏感性。Objective To analyze the effect of targeted silencing S100A4 gene on cisplatin sensitivity of endometrial cancer cells.Methods Virus transfected endometrial carcinoma KLE cell lines and obtained stable cell lines.The expression of S100A4 mRNA in transfected endometrial cancer cells was detected by polymerase chain reaction.The expression of S100A4 protein in endometrial cancer cells was detected by Western blot.CCK8 assay was used to detect the proliferation of endometrial cancer cells after transfection.Results The optical density values of the negative control group,the shRNA interference group and the overexpression group were(0.211±0.057),(0.216±0.034),(0.204±0.058)and(0.282±0.083),respectively.The expression level of S100A4 protein in the blank control group and shRNA interference group was significantly lower than that in the negative control group,and the expression level of S100A4 protein in the overexpression group was significantly higher than that in the negative control group,with statistically significant differences(P<0.05).The IC50 values of blank control group,negative control group,shRNA interference group and overexpression group were(118.400±0.021)nmol/L,(102.600±0.021)nmol/L,(89.700±0.021)nmol/L and(121.500±0.018)nmol/L,respectively.Conclusion Targeted silencing of S100A4 gene can inhibit tumor cell proliferation,promote cell apoptosis,and enhance the sensitivity of endometrial cancer cells to Cisplatin.
