摘 要:【目的】调查不同氮肥水平处理对水稻OsAAP6基因表达的影响,并进一步探究OsA4P6基因在不同氮肥条件下对其水稻籽粒中蛋白质(包括谷蛋白、醇溶蛋白、球蛋白和清蛋白)含量和氨基酸含量等营养品质的影响作用,为今后利用OsA4P6基因提高稻米营养品质提供参考。【方法】设置5个氮肥浓度梯度,在水稻OsA4P6遗传材料中分别检测OsA4P6基因的表达水平,并利用近红外谷物分析仪和考马斯亮蓝G-25O法检测分析其成熟种子中蛋白质含量和氨基酸含量等稻米营养品质性状。【结果】随着田间氮肥施加量的增加,OsA4P6基因的表达水平明显提高,并能显著促进水稻籽粒中谷蛋白的积累,进而增加水稻籽粒中总蛋白质的含量;在氮肥浓度较高的条件下基因有助于提高水稻籽粒中总必需氨基酸的含量;随着氮肥施加量的增加,OsA4P6基因能够显著提高水稻籽粒中总氨基酸的含量。【结论】OsA4P6基因随着田间氮肥浓度的增加,其表达水平不断升高,并能促进水稻籽粒中蛋白质含量和氨基酸含量等稻米营养品质的提高,这为今后利用OsA4P6基因提高稻米的营养品质提供了重要参考。【Objective】This paper aimed to investigate the effects of different nitrogen levels on the expression of 0sAAP6 gene in rice and further explore the effects of 0sAAP6 gene on the contents of protein(including glutenin,prolamin,globulin and albumin)and amino acids in rice grains under different nitrogen fertilizer conditions so as to utilize 0sAAP6 gene for improving the nutritional quality of rice in the future.【Method】Five nitrogen concentration gradients were set up to detect the expression level of gene from the genetic materials in rice,and the protein contents and amino acid contents in the mature seeds by near infrared grain analyzer and Coomassie Brilliant Blue G-250 method were analyzed.【Result】With the increase of nitrogen fertilizer concentration in the field,the expression level of 0sAAP6 gene in-creased significantly,which could promote the accumulation of glutenin in rice grain and then increased the total content of grain protein in rice.Simultaneously,with the increase of nitrogenous fertilizer application,0sAAP6 gene could significantly increase the total content of amino acids in rice grain.【Conclusion】gene expression level increased with the increase of nitrogen fertilizer concentration in the field,and could promote the nutritional quality in rice,such as the protein content and amino acids content in the grain,which provided an important reference for the future utilize of 0sAAP6 gene to improve the nutritional quality of rice grain.
