摘 要:目的:研究T淋巴瘤侵袭转移诱导因子1(Tiam1)在膀胱癌组织表达与膀胱癌患者临床病理特征及预后的关系,并探讨Tiam1对膀胱癌细胞侵袭转移的影响及作用机制。方法:qRT-PCR和免疫组化检测Tiam1在70例膀胱癌组织和40例正常膀胱组织中的表达水平,qRT-PCR检测Tiam1在膀胱癌细胞系和人正常膀胱上皮细胞系中的表达;经脂质体分别转染Tiam1 siRNA(si-Tiam1)和对照(si-NC)48 h后,采用boyden和Transwell实验分别检测各组细胞侵袭和转移能力,Western blot检测各组细胞中MMP7蛋白的影响。结果:qRT-PCR和免疫组化结果显示Tiam1在膀胱癌组织中的表达显著高于正常膀胱组织,其高表达与肿瘤临床分期和淋巴结转移显著相关(P值均<0.05),膀胱癌组织中Tiam1高表达患者生存期较短。Tiam1在膀胱癌细胞系中的表达均显著高于人正常膀胱上皮细胞系SV-HUC-1,其中T24细胞中Tiam1的表达最高(P<0.05);转染Tiam1 siRNA后,T24细胞侵袭转移能力下降、MMP7蛋白的表达减少(P值均<0.05)。结论:Tiam1在膀胱癌组织和细胞系中均高表达,且高表达与临床分期、淋巴结转移及不良预后相关,同时Tiam1可能通过作用于MMP蛋白促进细胞的侵袭转移,Tiam1可以作为侵袭转移膀胱癌患者的不良预后分子及治疗膀胱癌的潜在靶点。Objective:To detect the expression of T lymphoma invasion and metastasis inducing factor 1(Tiam1)in bladder cancer,to explore its clinical significance and prognosis of patients with bladder cancer,and to investigate its effect on invasion and metastasis of bladder cells.Methods:qRT-PCR and immunohistochemistry were used to detect the expression of Tiam1 in 70 cases of bladder cancer and 40 cases of normal bladder.qRT-PCR was used to detect the expression of Tiam1 in bladder cancer cell lines and human normal bladder epithelial cell lines.After transfection with Tiam1 siRNA(si-Tiam1)and control(si-NC)for 48 h,the boyden and Transwell experiments were performed to detect the invasiveness and metastasis ability of each group cells,and the expression of MMP7 protein in each group cells was detected by Western blot.Results:qRT-PCR and immunohistochemistry showed that the expression of Tiam1 in bladder cancer tissues was significantly higher than that in normal bladder tissues.The high expression was significantly correlated with clinical stage and lymph node metastasis(P<0.05).Patients with high expression of Tiam1 had a shorter survival period.The expression of Tiam1 in bladder cancer cell lines was significantly higher than that in human normal bladder epithelial cell line SV-HUC-1,and the expression of Tiam1 was the highest in T24 cells(P<0.05).The invasiveness and metastasis ability of T24 cells and the expression of MMP7 protein was decreased after transfected with Tiam1 siRNA(P<0.05).Conclusion:Tiam1 is highly expressed in bladder cancer tissues and cell lines,and high expression is associated with clinical stage,lymph node metastasis and poor prognosis.Tiam1 may promote cell invasion and metastasis by acting on MMP protein.Tiam 1 can be used as poor prognostic molecule for invasion and metastatiasis bladder cancer patients and potential target for the treatment of bladder cancer.
