摘  要:目的探讨miRNA-145在膀胱癌侵袭和转移中的潜在调控作用及其分子机制。方法采用反转录-定量聚合酶链反应法检测miRNA-145在膀胱腺癌组织和膀胱癌细胞中的表达。将miRNA-145模拟物和抑制剂转染到膀胱癌(BC)细胞中,通过伤口愈合和侵袭检测,确定miRNA-145对细胞活力和侵袭性的作用。结果与健康对照组相比,在有迁移和侵袭BC患者的细胞和组织中miRNA-145明显下降(P<0.01)。结论miRNA-145明显抑制了BC细胞的迁移和侵袭能力。Objective The current study aimed to investigate the potential modulation of miRNA-145 in bladder carcinoma and elucidate the underlying molecular mechanism.Methods The expression of miRNA-145 in bladder adenocarcinoma tissues and bladder cancer cells was detected by reverse transcription-quantitative polymerase chain reaction.MiRNA-145 mimics and inhibitors were transfected into bladder cancer(BC)cells to modulate the expression of miRNA-145.Through wound healing and invasive testing,we determined the effect of miRNA-145 on cell viability and invasiveness.Results MiRNA-145 decreased significantly in cells and tissues of BC patients with migration and invasion compared with healthy controls(P<0.01).Conclusion MiRNA145 markedly inhibits the ability of BC cells to migrate and invade.The results of the present study indicates that miRNA145 could function as a tumor suppressor and might have a potential to be a diagnostic and predictive biomarker,as well as a therapeutic target for treatment of BC.
